Larry, of Leisure Suit Larry fame.

"Meet Larry. He's just turned 40, and he's still single. If you can get past the glow of his Grecian formula, you can see his hairline beginning to make a hasty retreat from his forehead. Larry's leisure suit is of the highest quality (100% manmade material, permanent press too!). He wears at least 11 gold chains and his freshly-capped teeth could blind you in a bright light. Down at the singles bar he tells all the chicks, "Sure, I'm single... I got in a fight with my ol' lady and she threw me out." He doesn't tell them the "ol' lady" was his mom, or that he was 38 at the time."

(Excerpt taken from http://adventure.if-legends.org/Sierra_On-Line,_Inc/Larry.html)

Ristar

Had he come out before Sonic, he would've made for an adorable mascot (ironically, Ristar was made by Sonic Team).

(extends arms in my direction)
"I wanna hug you..."
'Aw, okay.'
(grabs me and constricts arms, ramming his head into me and sending me flying)
"Ha ha."
'Oww... I forgive you, though; you're just too cute to hate.'

And I have no clue what to call this Second Life avatar someone created. It's rather disturbing and arousing at the same time o_O

If we're talking about characters who appear in more than one game, then I'd have to go with Travis Touchdown.

I picked Travis mainly because he's really obscure, and not an ordinary video game character. He's a nerd, and he's proud of it. That, and he could be a decent representative for gaming, if he weren't so bloodthirsty and arrogant.
This. Travis kicks arse.

Waluigi clearly deserves a mention. His name is absolutely ridiculous in American, and a clever pun in Japanese. That skinny bastard is a great alterna-mascot.

How about the Fight music from early Pokemon for the Gameboy Colour, and the Finaly fantasy fight music, I used to watch my friend play that when I was a kid, it drove me insane.

My absolute favorite is Zyzyx from my favorite game, Sacrifice. That wise-cracking, sarcastic, know-it-all lil' imp just warms my heart. Especially when he helpfully cries out "I KNEW IT WAS A TRAP!" about five seconds after the fact...

Minsc and Boo! [http://www.minscandboo.com/index.php]

How can you not like a half-crazed but oh so lovable and rightgeous Barbarian Ranger with his Giant Minature Space Hamster companion, whom imprint their boots of justice into the swollen and unsightlky backside of Evil, anywhere it'd hides!?!!
Seconded.
"Evil around every corner. Make sure not to step in any."
Third for Minsc, the defender of goodness. "Make way, villainy! Hero, coming through!" Such a classic

I believe right now it's a tie between Minsc and Boo (they are a single entity ... *glares*), the Companion Cube (excellent mascot, very minimalistic and Art Deco and all XD) and I'm not so sure if the comments count as votes but that John Vattic guy might be in the run against them too!

I have lost myself in your words, but no matter!
Worth the price of the game all by itself.

This one isn't my favorite but its in the top ten: Mudcrabs from Oblivion.
Seriously, every Oblivion player knows about mudcrabs and I can't think of Oblivion without thinking about mudcrabs and vice versa.

The Security Guards from Half Life. My heart really went out to them when I saw one pathetically firing at a Gargantua, moments before he was slammed by a car and crushed into a wall. His last words were, "Gotcha!"

He died as he lived.

I'd say April Ryan, the staple character of The Longest Journey franchise and possibly one of Funcom's most iconic characters. April is still one of the most well written and well acted characters I've seen in the videogame genre, if not entertainment in general. She has real character development and a personality that isn't taken from the usual cookie cutter mold that most female game characters are in this day and age.
