I'm probably going to go with... Either Steve Hughes or George Carlin
It took 18 comments for someone to mention George Carlin... I am severely disappointed.

OT: George Carlin. No contest.

If there were runner up positions, though, they would go to:
2. Brian Regan
EDIT: Actually, tied for #2 is Robin Williams.

3. Jim Carrey
Unfortunately that's his only television-produced stand-up show that I've ever been able to find. He has a few clips circulating here and there, but that's the only full show I've heard of.

4. Christopher Titus

Mine is/was Mitch Hedberg. I loved the non sequiturs, and how he did the whole stand-up comedian thing wrong. There are many things in life that should be ho-hum to me, but there's a Mitch joke behind them that makes them wonderful.

Sammy J and Randy.

They are brilliant.

I can't be bothered linking, just look 'em up.

Ever? George Carlin, by far. The man was a genius.

Currently it's Louis C.K. A spiritual successor to Carlin in every way.

Christopher Titus gets an honorable mention too.

I'm torn between Billy Connolly and Gabriel Iglesias. Ross Noble and Akmal have great moments too (don't let Noble near a smoke machine unless you want the whole show derailed).

Can't narrow it down to one, so on a scale going from 'scripted and structured, but intelligent' to 'improvised and random, but slightly mad': Dylan Moran, Dara O'Briain, and Ross Noble.

Don Rickles. Too bad he won't be around much longer. The OG bad boy of comedy who didn't give a fuck who he insulted, but it was so goddamn funny they couldn't help it. He's so funny, even the inside jokes are hilarious.

I am a firm supporter in the holy trinity that is Kinnison, Carlin and Hicks, RIP.

Though... one who is still living who merits an equal level of respect is Titus.

Anyone here who has not seen one of Jim Jeffries's cramp-inducing routines needs to stop whatever worthless bullshit they're currently doing and take ten minutes to watch this right now. That's an order.
Seriously. Watch it, god damn it.
I'm not kidding. You will thank me.

If that video won't make you laugh out loud, you're a wrong person.
I couldn't breath. At first I didn't think he was gonna be funny, but...as it went on I just couldn't stop laughing.

Christopher Titus, no contest. He's fucking hilarious and I find him very relatable...though that may say something horrible about me and my family.

Here's a couple of my favorite bits.

Everyone can go home now! The thread is over.

Seriously, no comedian can even come close to Christopher Titus for me. I'd post some more videos (because who can really have too much Titus?!), but I'm on my phone now, so that would be difficult.

Other notables though include Chris Rock, Daniel Tosh, Jim Jefferies, and the late, great Mitch Hedberg. This probably rounds out my top five, though after Chris Rock, I don't know 100% about the order.

its really hard to choose because there are so many but i would have to say frankie boyle because he crosses those lines no one else dare cross, because he is not the comedian we need but the comedian we deserve, a comic warrior, a staunch protector, a dark knight.

seriously with comedy becoming more and more PG and people complaining more and more about "offensive" jokes and taking more and more backlash for being rude we need people like frankie boyle and to defend comedians rights to explore controversy and make rude and "offensive" jokes
His stuff on MTW was excellent, but I've always found his stand-up pretty shit. He works well on panel shows.

Anyway, currently working? Dara O' Briain. Saw him live quite recently, and he was hilarious.
Of all time? Difficult to choose, but I think I'd say Richard Pryor. I'm not old enough to have seen him when he was alive, but I've seen DVDs of a couple of his live shows, and my god, they are brilliant

Mike Birbiglia.

I want to post a video of his standup, but it's all long-form.

Hell, I'll post it anyway. It's like an hour, but the payoff is good for anybody who likes the more awkward Gaffiganesque humor.

Robin Williams.
I know a lot of people don't like him, but god damn I find him hilarious. He's batshit insane and works so fast you gotta watch his show three times to get every joke. He's got a huge variety too.

Russell Peters. This guy consistently cracks me up. This is an older clip but classic (although politically correct folks may want to skip it):
