I guess i'll say Megatokyo since no one has mentioned it yet. It had a severe drop in humor after the guy one of the characters is based on left but it's still worth checking out. I can't guarantee whether or not you'll enjoy it though.

EDIT: I remembered another good one called Aikonia, it's by the same person who does Awkward Zombie. It's actually has a story though, a damn good one at that. I'm really looking forward to find out what is going on.

its fantastic, good humour, although more mature and plot focused (theres a huge cast of characters and themes ranging from sexuality,relationships, oh and the main character is a gang leader.

Only one i hvnt seen mentioned here is grim tales. Its on a website called SNAFU and it brings in all ur childhood cartoon characters and gives them a wonderful dark and twisted slant

Aside from ones that have already been mentioned, Order of the Stick is pretty good and anything on Direman Press is a good laugh.

Most of the ones I read are already here, but here are some I didn't see:

anti-HEROES (an OoTS fan-comic, and yes that is the proper capitalization)

Servants of the Imperium (Warhammer 40k comic done in a similar style to OOTS)

The Noob Comic (about MMOs in general)

Goblins (of the D&D variety)

8-bit Theater (should not need any introduction)

Rusty & Co. (D&D, similar concept to Goblins)
