Vocals-Garrus Vakarian. You gotta admit he'd have one of the best voices ever for ballads.
Lead Guitar: Ezio Auditore da Firenze. Because that would be just plain awesome.
Bass-Shadow the Hedgehog. I dunno why, but he always struck me as a decent bassist.
Drums-The Demoman. Just because.
Keyboard-Sheogorath. Dude would come up with some of the trippiest keyboard parts since Pink Floyd.

OK, yesterday, there was a topic about who would make up your band if you pick and choose anyone in existence. Most people made their bands from real musicians, but I chose to list characters like Gadalf, Batman, and The Doctor, as well as some gaming characters.

Multi-instrumentalist: Leonardo Da Vinci (Assassin's Creed II). He does a bit of everything, right?

Alcatraz from Crysis 2 as lead vocals and guitar.
Isaac Clarke from Dead Space 2 on bass.
Sovereign from Mass Effect as the synths and transportation for the squishy organic lifeforms that make up the rest of the band. EDIT: And occasional vocals for the heavier songs, just to please the OP. ;)
Adrian Shephard from Half-Life: Opposing Force on drums.
Cole McGrath, because I imagine his lighting powers could make for some interesting miscellaneous sounds and awesome light shows for stage performances.

I'm imagining this being a Nine Inch Nails-y industrial metal band, only a billion times more awesome than that. Also, Alcatraz may seem like an odd choice for vocals considering the fact that he's silent in the game, but he does talk in this trailer, and his voice is awesome:
