The crowbar from Half Life, tap a couple times and they die...

Or Painkiller from Painkiller, I killed a boss with it and it couldnt kill me.

If it is by effect, Jumping on people's faces in Mario.

If it's just for being flashy, BFG from doom is a classic.

Blaster launcher from X-Com: Enemy Unknown. Not only does it cause a ridiculously huge explosion and more damage than any other weapon in the game, but the projectile can be guided around corners, into UFOs, up elevator shafts and through mazes. You don't even have to leave the transport craft to clear a map.
However, it is just as deadly in the hands of the aliens, and by late-game, half of them will be carrying one.

Joking aside, I really hate that gun and I avoid users who keep using it, I rather have my Gnasher battles on the Beta.
Haha, yeah i can see it being massively annoying (it's happened to me a few times obviously). The Gnasher i find to be a better overall weapon (well, from the amount of times my head seems to dissappear). More bullets before reload (as well as a quick reload), better range and is no less deadly at the same range as the Sawed-off.

The only advantage the Sawed-off has is you don't need to be so good at aiming, which is a bonus for me!

In minecraft:

Step 1: Make (or hack in) a lot of TNT, get some redstone and make some redstone repeaters.
Step 2: Cover an island in it. For bonus points, dig down about 20 blocks and fill that with TNT as well.
Step 3: Wire a detonator and prepare a safe place to watch the carnage. This should be very high so you can watch your island disappear. You should be able to set the explosion from this safe place.
Step 4: Prepare maniacal laughter.
Step 5: Press the button.
Step 6: Watch the madness. Don't be concerned if you take a sizable performance hit - this is normal during massive TNT explosions.

The Cain (AKA Nuke Launcher) in Mass Effect 2. It's so powerful I've never had an occasion when I actually needed to use it. And I was scared to use its single shot per mission (sans upgrades)...

Why wouldn't I. Watching 8 mini-nukes slowly approach someone, start to smash their face and explode violently is hilarious.

I too have lauched myself off of Tenpenny tower. I don't remember the game froze or not.

M-98 Widow from ME2. Fully evolved Infiltrator with assassination cloak and tungsten ammo, will take down a YMIR mech in 2 shots or Harbinger in 1. May not be as massive overkill as some of the others on this list, but against run of the mill opposition... splut...

Plus it requires a bit of old-fashioned craftsmanship to use :D

The rail driver (Red Faction). The scope allows you to see enemies' heat silhouettes through all manner or walls or terrain, and the slug it fires can penetrate anything at any distance. And it's a guaranteed one hit kill, but the reload is slow and one slug can be fired at a time. Snipers are better off with the actual sniper rifle in the game since it can basically do the same thing, without the benefit of wall penetration.

The Bring it on Home solo from Brutal Legend... effing awesome, especially when you time it right and it crashes down into your enemy's army right at the start of a battle. Or when you're about to take down their stage and you play it, and as you watch the stage collapse a burning blimp just flies down and crashes into it.
