In no particular order and based around how much fun I had rather than how much effect it had on the industry or anything.

Crusader Kings 2
Guild Wars 2
Civilization V
Mass Effect 3
Valkyria Chronicles
Armed Assault 2 (for me anyway)
Dragon Age: Origins
FTL: Faster Than Light
Dynasty Warriors: Gundam 3
Alice the Madness Returns

Timesplitters 3: Future Perfect is still the best game ever made though.

Civ 4 was technically last generation. It came out October 05. The 360 came out November 05. Thus Civ 4 is one of the last games of the previous generation. Sad as it would have made my list easily if not for this.
Okay... ahem:
4) Civ 4 WITH Fall From Heaven 2... that came out around May of 06. Wheee! :p

1) Starcraft II
2) DotA 2
3) World of Warcraft
4) VVVVVV
5) Super Meat Boy
6) Gundeadli Gne
7) Dark Souls
8) Skyrim
9) Rayman Origins
10) Street Fighter IV

One could argue that WoW is too old to be "this generation" and DotA too new. Both being PC games I don't think it's relevant tough.

Not sure if a mod makes it count but whatever. Just glad to see someone else still enjoy this excellent game. ^_^

And don't get 5. It is crap. 1 unit per tile, extremely dumbed down diplomacy, laggy, buggy, many civs kept separate to be sold as DLC not in expansions.
WHAT?! I can see why they would try to limit, for realism, but 1 unit? At least make it two at a time so I can set up a defensive wall and strike through it, I'd even pull back the striking unit after hitting once. The civs as dlc thing is bad enough, though. How come FPS players didn't put up with that crap when their guns were being sold as dlc, but we're willing to put up with it with our player choices? Finally, the diplomacy is Civ 4 is already plenty dumb, how'd they make it even simpler?

As far as a generation go, that means anything released in the last 7 years or so right?

1- Dwarf Fortress (2006)
2- Crusader Kings 2 (2012)
3- Medieval 2: Total War (2006)
4- STALKER: Call of Pripyat (2010)
5- Company of Heroes (2006)
6- Galactic Civilizations 2 (2006)
7- Dawn of War 2 (2009)
8- Mass Effect series (2007-2012)
9- Neverwinter Nights 2 (2006)
10- Mount & Blade: Warband (2010)

Honourable mentions:

-Bastion
-Witcher 2
-Telltale's Walking Dead
-Arkham Asylum/City
-Starcraft 2
-Dragon Age: Origins
-Civilization 4 (Technically came out 1 month before the 360 so didn't count it)
-Tropico 3/4
-Telltale's Sam & Max series

1: New Vegas
2. Portal 2
3. Bioshock Infinite
4. FIFA 11
5. Modern Warfare 2
6. Skyrim
7. CoD4: Modern Warfare
8. Xcom Enemy Unknown
9. Fallout 3
10. Red Dead Redemption

Some odd choices in there, but the CoDs, Portal 2 and FIFA 11 were some of the best multiplayer experiences I've ever had. Not for balance or anything, but for a fair few months most of the nights I came back from 6th form me and 4-10 of my mates would hop on Xbox and play one of the Cods or FIFA. And laughs and fun were had every time, without fail. The anger you feel at the problems in game kind of go away a chunk when you're playing with friends. Portal 2 co-op I played with one of my good friends and it was fun, hilarious and puzzling. A great bit of time spent there.

In terms of those that defined this generation (if you did a montage tribute to it these would be prominent):

Gears of War, CoD:MW, Halo, Assassins Creed, Dead Space, Batman: Arkham, Boarderlands, Wii Sports and arguably I think the Mass Effect series and of course the game changer that is Minecraft.

For me personally (as in when I look back what I'll remember the most, or if I made a montage this is what it'd be).

1. Skyrim
2. Fallout 3
3. Mass Effect series
4. The Walking Dead (eps 1-5)
5. Assassins Creed series
6. Batman: Arkham series
7. X-Com: Enemy Unknown
8. Dead Space 1 & 2 ('cus fuck 3)
9. Crysis (just the first)
10. Just Cause 2

Admittedly most of these I played on PC so the generation thing doesn't really apply. Maybe I'll do another one later just for consoles.

10) Mass Effect
9) Metroid Prime 3: Corruption
8) Batman: Arkham City
7) Limbo
6) The Walking Dead
5) BioShock
4) Portal 2
3) Batman: Arkham Asylum
2) Minecraft
1) Portal (or the entire Orange Box, if that counts [which it probably shouldn't]; regardless, Portal, being my favorite game of all time, is still more than enough to take the top spot on its own)

Let it be noted that I unfortunately have yet to play a large number of the games that populate most of the previous lists, so the exclusion of many titles like Skyrim, Spec Ops: The Line, Dark Souls, Bastion, Mass Effect 2, BioShock Infinite, etc. is likely thanks to my own deprivation rather than a purposeful desire to exclude them from my list.

I'm going to preface this by saying I don;t own a PS3 (I know I'd love Disgaea 4 and I haven't played Uncharted, for example), and leaving out PS2 games released in this gen (Persona, Dragon Quest VIII, etc.) And also, no order, and no two games in the same series

Ok, here we go...

Dragon Age: Origins
Mass Effect 2
Civilization V
Metal Gear Solid 4: Guns of the Patriots (one of the only PS3 games I got to play)
Batman: Arkham Asylum
Pokemon Black 2/White 2 (SoulSIlver/HeartGold are absolutely astounding, but Black 2/White 2 get more credit for being completely new)
Super Mario Galaxy 2
Phoenix Wright: Ace Attorney
Advance Wars: Dual Strike
Shin Megami Tensei: Strange Journey I love Devil Survivor, I just... like this a bit more, maybe because it's more not typical Shin Megami in setting but still pulls it off just as well)
