I think post American civil could work quite well, the excuse could be they're finding an ancestor with the best gun skills for Desmond.

You'd have the Assassin's blades for quiet work. Two revolvers on your belt, these would be loud with limited ammo but eventually become really good for clearing big groups of enemies. You'd get a rifle or shotgun slung over your back (a choice between long range versus close heavy power). You'd also have a melee weapon ranging from sabres, bowie knives and tomahawks.

The Assassin would wear a white great coat over his clothes and wear a white hat (these you could just like AC2).

The cities could be an early New York (in role of Florence in AC2), you could climb building without them being too tall. The last city could be New Orleans (fulfilling the Venice role) and the two smaller towns could be Dodge City and Tombstone (for the cliches).

Imagine running alongside the rooftops of a ramshackle western town as guards on the ground fire lever action rifles. You leap across a gap, on to some hanging scaffolding, pivoting as it rocks through the air and firing two pistols back at them.
