Exactly what the title says, not what is the best game, but what single act in a game just makes you feel fuzzy or awesome or epic, whatever you want. For me it has to be getting the PERFECT block in Tetris, the gratification is so frequent and simple but it never dulls.

Hitting 80 in wow was pretty awesome. Even if you hate the game, think of the equivilent in your MMORPG/RPG of choice.

Getting a long unbroken moment of badassness in a shooter, when you get that blessing of luck and suddenly you are taking down hundreds of enemies all the while dodging their attacks and you are in complete concentration because if you break it for even a second you will die and you sort of forget where you are because all that exists is this badassness and man the adrenaline is pumping like mad, Halo, Call of Duty, Half Life 2, Team Fortress 2, Gears of War... any game like that... it makes me think: "OH GOD, I'M ALIVE!!!"

Exploring. It's that feeling of total freedom you get upon reaching the top of a high point in the landscape and looking out over the panoramic view, just you and your character. Especially if getting to that high point involves jumping all over the town like a medieval yamakaze master. =3

Being at a point in Oblivion where I can do anything I want.

Of course trying out new cool powers and abilities in almost any game is pretty cool too.

Certain game endings with good music can feel quite nice. I also get a warm, cozy feeling from sniping from a nest.

I love the feeling of a new game, that exitment you feel when seing the forests of Cyrodiil for the first time, the rush you get when the first zombie charges you in half-life 2, The laugh you get when crashing a car in burnout paradise and the thought when sniping someone on team fortress 2, and of course the amazment when Alex mercer is being a dick, killing random civilians. that is the best feeling of gaming, the feeling of somthing new.

Watching an enemy's head exploding in slo-mo.
Or watching them get ripped in half by sniper fire. Not in slo-mo.

It was like beng tortured for three hours then given a lovely massage...

A feeling of pure relief and joy, yet somehow still traumatised

Hearing the announcer shouting "Q-q-q-quad kill!" followed by "Genocide!" and finally "Champion of Newerth!" in Heroes of Newerth (dota with better graphics) almost never gets old... especially since i got it with a support class character.
