Honorable mention goes to the end of Cowboy Bebop, which sadly gets disqualified for not being a movie.
First comment drops one of the most epic...well played, man, well played indeed. I'll continue with this theme of destruction by dropping in V for Vendetta and Gran Torino. "Now me...I got a light. It's right here." What a way to top it off. On a side note, the best movie to display glee and delight without ever shoehorning in an annoying and unnecessary love plot (no, really, there isn't one! Gasp, I know!) is Mr Magorium's Wonder Emporium, and gets a bit of a runner-up medal for having a damn fun ending.
I really liked that movie until the ending. The ending made me dislike the movie overall, because the main character's behavior was in stark constrast to everything he'd done over the entire film. I found it to be silly, but to each their own, I guess.
That is not the ending. The ending is a monologue by Sam Elliot, and it's awesome.
The dude abides... indeed. I don't know about you, but I take comfort in that: That he's out there, "The Dude", taking 'er easy for all us sinners.
OT: The Usual Suspects, Twelve Monkeys, the Departed, and Dr. Strangelove are all good. I'll even throw another relatively new one in there: Thank You for Smoking.
All these pale in comparison, though, to the greatest ending of all time: One Flew Over the Cuckoo's Nest. I'm serious. If you haven't already, go see it. Right now.
I would agree with you on Silence of the Lambs... *chills*
Buuut I prefered the ending of The Life of Brian. Monty Python songs are always awesome!

Um... hmm... I can't think of anything that hasn't already been mentioned. I'd like to mention LotR: Return of the King, if only because they stayed truer to the book than I would have thought feasible for a Hollywood-type film.
The end of the third series of The League of Gentlemen was probably my favourite ending to a comedy series ever, I was amazed they managed to wring so much emotion out of such an unpleasant cast of characters.
And if we're talking about anime, I loved the ending of Neon Genesis Evangelion. And I mean the original ending. So they had budgetary and time restraints; IMO that just forced them to be more creative and abstract, something which was ultimately to the benefit of the show.
Semi-visible sex scene in a stasis tube FTW. With the President of the International Federation demanding to see them within 20 seconds.
And epic music.

The ending of Moulin Rouge is one of my favorites.
It's the end of the performance, and you almost for a few seconds forget that it's a tragedy. Then Satine dies, and you're going "but she's gonna come back, right? Right?" Nope curtains close. Sad ending.

Well, besides fight club (Damnit...) I'm going to have to go with DayBreakers
i mean, it ended with the opening for a part 2, but the whole turning the boss back into a human just to have him get chomped to death by his underlings was just great.

unfortunately there might be a new series because the author said "Spike only did what the person thought, he might of died or just past out... who knows?"
Okay, sorry for doing this, but I am sick of and tired of this mistake. IT'S "HAVE" NOT "OF"! IT'S NOT "SHOULD OF" (OR IN THIS CASE, "MIGHT OF") IT'S "SHOULD'VE" (MIGHT'VE), WHICH IS AN ABBREVIATION OF "SHOULD HAVE" (MIGHT HAVE)! WHEN ARE PEOPLE GOING TO GET THIS RIGHT?
OT: I second Holy Grail, it comes out of nowhere, but it isn't completely random.
