I don't think he was always a religious man, but something happened to him, war hero perhaps? Whatever it was, he seeked out religion after the fact. He knows too much to have always been a clergyman.
Whedon confirmed in some thingy that Book was a spy (cant recall if he was spying on or spying for the Alliance - google would help if you're keen) and that he got stupidly high within the ranks of the Alliance. However he killed a shepherd one day and felt so guilty that he took on that man's identity to remake himself.

Lots of hints are dropped throughout Firefly to this of course like his knowledge of guns and Alliance tactics and structure. Most handed clue of course is when Early knocks him out and says to Simon, "Thats no shepherd.".

Gandalf, also known as Olórin in his youth in the West. I know that he technically shouldn't count and that Tolkien didn't really want any religion in his books but this here, it's just... holy, that's all I can say. What man could be more holy, profound and awesome (in the archaic sense) than Mithrandir, the Grey Pilgrim?

Okay, so fair's fair, Imperial Redemptionists aren't exactly nuanced. There are still some pretty interesting ideas bouncing around in the Imperial faith, especially the fact that in the Imperium worshipping the Emperor can take almost any form imaginable.

Buuut I think the award goes to Dsnte Aligeri because not only did he write a long ass poem, he stabbed Genghis Khan in the face with death's scythe and murders demons by crushing there skulls with a blessed crucifix that fire giant crucifix silhouettes.

Honestly? I love The Arbiter. He was devout to his religion and carried it with honor. At the same time, however, he was somewhat open to new ideas and at least listened to Guilty Spark. Even when he had his whole faith destroyed, he stayed calm and found something to keep him going.
