Nah, I've never really seen anything close to a "best" character yet. Besides, it's your own subjective opinion based on how much you can identify with the character.

But I have to say I really like Bill from L4D.

I'm going to be original (and cheat by choosing two characters) and say Monkey and Trip from Enslaved:

Both very interesting characters, especially when they're together. Andy Serkis isn't in games very often, but he pulls of the role of Monkey quite fabulously. Same goes for Lindsey Shaw for Tripitaka.

Ninja Theory went all out on this one.
