Right, here's the immediate and obvious thoughts:

Mario Kart DS
New Super Mario Bros.
Nintendogs
Pokémon
GTA: Chinatown Wars
Animal Crossing
Professor Layton
Chrono Trigger

And here are the slightly less obvious:

Hotel Dusk
Phoenix Wright
Elite Beat Agents
Bleach: The Blade of Fate
Custom Robo Arena

Etrian Odyssey 2 & 3

You can probably skip EO1, since EO2 is, more or less, an upgraded and class balanced version of EO1.
EO3 continues upgrades to the gameplay and uses entirely different classes than EO1 & EO2.

Legend of Kage 2:
It's a slash-em-up with some RPG and magic elements.

It's already been said, but Pokemon. I'm not kidding; you'll get far more out of it than you might any other DS game.

Seriously? Only two other people have mentioned The World Ends With You? If you aren't into pokemon thats seriously the only game I would recommend someone getting a DS. I'm on my third play through at the moment.

Combats super fun. The story line is decent for something coming from Square Enix. The visuals are sweet. It seriously makes use of the DS's every feature. Pokemon could just be played on any old console.

If you had money to burn I might also suggest GTA:Chinatown, if you didn't mind pinballing down narrow streets and cars. I had fun just jumping from dealer to dealer selling drugs as if it was some fucked up stockmarket where cops could suddenly bust in on the party to say hi.

Scribblenauts was fun for a laugh as well.

Elite Beat Agents was good if you have a rhythm game itch to scratch. The difficulty curve is intense if you wanted a challenge.

Geometry wars was fun while it lasted as well, I got on one of the top 100 scoreboards for that once. Apparently you can network your DS with your Wii on this one and do multiplayer matches, I'm not quite sure how that works though, since I never owned a Wii.

Besides Pokemon and TWEWY, I would HIGHLY recommend Luminous Arc 1 & 2, the story is great and the gameplay is reminiscent of Disgaea but a bit less organised and WAY less forgiving. Luminous Arc 2 got a mention in an article a while back, which is how I learned of its existence:
http://www.escapistmagazine.com/articles/view/issues/issue_279/8296-Wussy-RPG-Girls.3
There's a short description of the game in the third paragraph.
All the Disgaea games are worth a mention too, hundreds of hours of fun out of any of them, oh, and there are Prinnys too. Prinny's make anything awesome, dood.

Phantom Hourglass is pretty good, DragonQuest IX is cool...Professor Layton if you like the puzzles is really good.

Osu! Tatake! Ouedan! is better than Elite Beat Agents but both are fun.
New Super Mario Bros is really good and Mario Kart on the DS is my favourite in the series.

Shin Megami Tensei: Devil Survivor
Shin Megami Tensei: Strange Journey

That way you can also see which game franchise Nintendo had to rip off to create Pokémon.

Pokemon Black/White are exceptional and surprisingly complex RPGs with a ton of variety.

Either of the Advance Wars are great.

Castevania Dawn of Sorrow - Haven't played any of the others, but DoS was my first Castevania game, and it's one of my favourite games on the system.

Wario Ware Touched. Wario Ware is a fun little series, and the touch screen lends itself very well to the conept.

Animal Crossing Wild World. Good to jump on for 30/40 minutes every day.

Hope that points you towards something you enjoy.

Elite Beat Agents
Super Scribblenauts
Warioware: Touched! and/or Warioware: DIY if you're into game-making or music composition
Civilization:Revolution
Advance Wars (either)
Cave Story (if you have a DSi)

New Super Mario Bros, Starfox, Bomberman, Warioware, Jam Sessions, Rhythm Heaven, Chrono Trigger, Trauma Center, the Phoenix Wright series, Viewtiful Joe, Feel The Magic, and Brain Age.

And my very favorite game on the DS if you don't mind looking for an import is Osu! Tatake! Ouendan.

The main ones I would recommend are Elite Beat Agents (fun the first time, great replayability, requires sound), Ghost Trick (only playable once, but very unique puzzles and storyline), and the first three Phoenix Wright: Ace Attorney games (again, only playable once, but incredibly memorable, and a unique take on adventure-puzzle style of games. First game I've known where paying attention to the storyline pays off)
