Mass Effect's "We held the line!" speech in Virmire.
Effective and simple.

For he today that sheds his blood with me shall be my brother; be never so vile.
This day shall gentle his condition.
And gentlemen in England now abed shall think themselves accursed they were not here,
and hold their manhoods cheap whiles any speaks that fought with us upon Saint Crispin's day!

The first instance I'm aware of comes from Henry V in the famous St Crispin's Day Speech:

"
KING. What's he that wishes so?
My cousin Westmoreland? No, my fair cousin;
If we are mark'd to die, we are enow
To do our country loss; and if to live,
The fewer men, the greater share of honour.
God's will! I pray thee, wish not one man more.
By Jove, I am not covetous for gold,
Nor care I who doth feed upon my cost;
It yearns me not if men my garments wear;
Such outward things dwell not in my desires.
But if it be a sin to covet honour,
I am the most offending soul alive.
No, faith, my coz, wish not a man from England.
God's peace! I would not lose so great an honour
As one man more methinks would share from me
For the best hope I have. O, do not wish one more!
Rather proclaim it, Westmoreland, through my host,
That he which hath no stomach to this fight,
Let him depart; his passport shall be made,
And crowns for convoy put into his purse;
We would not die in that man's company
That fears his fellowship to die with us.
This day is call'd the feast of Crispian.
He that outlives this day, and comes safe home,
Will stand a tip-toe when this day is nam'd,
And rouse him at the name of Crispian.
He that shall live this day, and see old age,
Will yearly on the vigil feast his neighbours,
And say 'To-morrow is Saint Crispian.'
Then will he strip his sleeve and show his scars,
And say 'These wounds I had on Crispian's day.'
Old men forget; yet all shall be forgot,
But he'll remember, with advantages,
What feats he did that day. Then shall our names,
Familiar in his mouth as household words-
Harry the King, Bedford and Exeter,
Warwick and Talbot, Salisbury and Gloucester-
Be in their flowing cups freshly rememb'red.
This story shall the good man teach his son;
And Crispin Crispian shall ne'er go by,
From this day to the ending of the world,
But we in it shall be remembered-
We few, we happy few, we band of brothers;
For he to-day that sheds his blood with me
Shall be my brother; be he ne'er so vile,
This day shall gentle his condition;
And gentlemen in England now-a-bed
Shall think themselves accurs'd they were not here,
And hold their manhoods cheap whiles any speaks
That fought with us upon Saint Crispin's day.
"
Spectacularly performed by Kenneth Branaugh:

This particular speech is the inspiration behind most epic pre-battle speeches including Braveheart, Independence Day and and so forth. Immitation is so common that such speeches are called "Saint Crispin's Day speeches".

In Mass Effect where Captain Kirrahe makes his speech of "holding the line"
Oh yeah, that was inspiring and honestly the stuff of legend.
I hate myself so much for screwing up on my main Shepard because I accidentally let Kirrahe die, I regret it sooo much.

Also on Mass Effect's topic, Shepard's speech just before leaving the Citadel was pretty inspiring, even better given that I made it myself.

Rome: Total War had most of the best game speeches. Particularly among crazy generals.

The enemy is furious today because I ate ... all ... their ... bees! They haven't had honey in many moons.

We will win today because our sharp pointy things are sharper than their sharp pointy things.

The speech before the Yunalesca fight in Final Fantasy X is the best pre-battle speech in gaming imo.

I'm not sure if it actually fits what you need, but the script's in the spolier.

Yunalesca: Have you chosen the one to become your fayth? Who will it be?
Yuna: Might I ask something first? Will Sin come back even should I use the Final Summoning to defeat it?
Yunalesca: Sin is eternal. Every aeon that defeats it becomes Sin in its place... And thus is Sin reborn.
Tidus: So that's why Jecht became Sin.
Yunalesca: Sin is an inevitable part of Spira's destiny. It is neverending.
Wakka: Neverending? But...but...if we atone for our crimes, Sin will stop coming back, ya? Someday, it'll be gone, ya?
Yunalesca: Will humanity ever attain such purity?
Wakka: Uh...
Lulu: This...this cannot be! The teachings state that we can exorcise Sin with complete atonement! It's been our only hope all these years!
Yunalesca: Hope is...comforting. It allows us to accept fate, however tragic it might be.
Tidus: No!
[Just as Tidus yells and charges, the ghost-image of young Auron reappears and does likewise, brandishing his sword]
Auron: No! Where is the sense in all this? Braska believed in Yevon's teachings and died for them! Jecht believed in Braska and gave his life for him!
Yunalesca: They chose to die...because they had hope.
[Young Auron gives a horrible yell and leaps, and Yunalesca zaps him. He flies across the room and crumples, then fades away.]
Yunalesca: Yevon's teachings and the Final Summoning give the people of Spira hope. Without hope, they would drown in their sorrow. Now, choose. Who will be your fayth? Who will be the one to renew Spira's hope?
Yuna: No one. I would have gladly died. I live for the people of Spira, and would have gladly died for them. But no more! The Final Summoning...is a false tradition that should be thrown away.
Yunalesca: No. It is our only hope. Your father sacrificed himself to give that hope to the people. So they would forget sorrow.
Yuna: Wrong. My father... My father wanted...to make Spira's sorrow go away. Not just cover it up with lies!
Yunalesca: Sorrow cannot be abolished. It is meaningless to try.
Yuna: My father... I loved him. So I... I will live with my sorrow, I will live my own life! I will defeat sorrow, in his place. I will stand my ground and be strong. I don't know when it will be but someday, I will conquer it. And I will do it without...false hope.
Yunalesca: Poor creature. You would throw away hope. Well... I will free you before you can drown in your sorrow. It is better for you to die in hope than to live in despair. Let me be your liberator.

Auron: Now! This is it! Now is the time to choose! Die and be free of pain or live and fight your sorrow! Now is the time to shape your stories! Your fate is in your hands!
Kimahri: Yuna needs Kimahri. Kimahri protect Yuna.
Rikku: Well, I'm fighting!
Wakka: I can't believe we're gonna fight Lady Yunalesca! Gimme a break!
Lulu: You can always run.
Wakka: Hah! I'd never forgive myself--no way! Not if I ran away now. Even in death, ya!
Lulu: My thoughts exactly.
Tidus: Yuna! This is our story! Now let's see this thing through together.

How about Gladiator? There was some good ones in there.

Or even Ben Hur. Baveheart. And I think Spartacus.

And of course anything from Shakespeare - which is really, as others have said, the main inspiration behind speeches said in most other films.

There are real life speeches too - such as Churchill, Kennedy, etc.

To be honest - I would go with Shakespeare then compare that against speeches made in real life and in cinema and show how Shakespeare's influence has stayed the course of time.
