Futurama, although the newer stuff seems to have lost something, like they're trying to ape Family Guy, thus totally missing the point.
I also love Venture Brothers (one of those rare cartoons that actually gets better as it progresses).
For defunct shows, i'd have to say Harvey Birdman: Attorney at Law (great from start to finish), Clone High (cancelled wayyyyy too soon), and Aqua Teen Hunger Force (yes, it's not technically defunct, but only the first 3 seasons were any good).

Not at all a fan of South Park (1/3 of the time it makes me laugh like i'm going to die, the other 2/3 it makes me want to puke), Family Guy, or American Dad. They can be funny, but they just bug me for some reason.

Not sure if its directed at kids or adults though, because anime has some weird shit directed at kids.
Great show. Definitely geared toward adults.

The Venture Brothers, is by far my favorite "adult" aimed American cartoon.
Yes South Park, Family Guy, The Simpsons, American Dad can be good for a chuckle once in a while be Venture Brothers is just so well done.

For me it's Futurama, South Park and Venture Bros i.e. the best show no ones watching.

EDIT Checking other posts it's nice to see at least some people are watching it.

In no particular order: South Park, Family Guy, American Dad, The Simpsons (let's say about 2001 and backwards), Futurama, Daria, Metalocalypse, Home Movies, King of the Hill, Dilbert, Clerks: The Animated Series (though I'm actually grateful it only lasted six episodes), The Boondocks and The Critic. If you counted anime i'd probably have a fair few more as well.

Daria, the original Ren & Stimpy (seriously, that is a @#$%ed up show), Home Movies, Sealab 2010...

...and MLP. Bwahahaha. Also, the original Looney Toons are just as great as ever, and if you think they're not for adults, then you obviously haven't watched them since you were a kid.

Actually, I think a lot of "kid" shows are secretly just big private jokes between a bunch of snickering animators and writers. Ever seen Yo Gabba Gabba? What's up with the big red cyclops dildo?

king of the hill, american dad, robot chicken, mission hill, tom goes to the mayor, the simpsons, south park(although now a days it's going down in quality), boondocks, metalocalypse, and aqua teen hunger force
there's still some I haven't mention but I can't think more

Invader Zim, It may be marketed as a kids show but from what i could see it was somewhat disturbing and funny so I do classify it as a "adult" show and a demented and funny one at that.

Venture Bros. I find that show so fucking funny. Other ones I like include Archer, Futurama, Metalocalypse, and South Park. I was about to put Robot Chicken in there, but then I realized it isn't a cartoon.

Although "Adult Cartoon" could be taken in an entirely different way...

Edit: Shit! Nearly forgot Ugly Americans. I will never forget that exploding lion...

Sealab 2021, Frisky Dingo, and Archer are all my favorite shows. I just love their conversational humor in a surreal premise.

There were a list related with "Best American adult cartoons" top 20 or something,
the number one was the Venture Bros,
and on that part there was a youtube video titled as "why The venture bros is the funniest show in 20 mins" or something.
I learned that show thanks to that site.
The show is really good and it resembles to the Tick - it was also a good superhuman cartoon parody. No not good, Perfect. Spoon, yes, spoon is a good thing.

Unfortunately, I couldn't find that site again...
