For the book(s) I have to go with the stereotypical whale choice, but not the monologue where he's naming things on his way to meet his new friend Ground. The description of the crater afterward had me laughing so hard the first time I read it I could hardly stand it. Of course I was about 11 at the time.

For the movie, it was hands down the first interaction with the infinite improbability drive.

Arthur: "Ford..."
Ford : "...yes..."
Arthur: "I think I'm a sofa."
Ford : "...I know how you feel."
Arthur and Ford: "........AHHHHH!!"

Lidrial - congrats on your 42nd post and thanks for a great topic that has made me smile warmly after the end of a really long and rough day.

For me, anyone who wins an award for their fjords is a prince in my book. And if his name is Slartibartfast and he wins awards for his fjords (that's fun to say - try it), then that man is a king.

I always have an honest answer when someone asks me what I think of their poetry - "Wow, this is so .... Vogon isn't it?"

I have two, off the top of my head. The first is when Arthur is stranded on prehistoric earth, and realizes after three years what his next plan should be.

Arthur: *declaring* "I shall go mad!"
Ford: "Splendid idea. I went mad for a while. Did me no end of good."
Arthur: *double-takes* "Ford?! I thought you were dead!"
Ford: "So did I, for a while. Then I thought I was a lemon. I spent weeks throwing myself in and out of a gin and tonic."
Arthur: "Where did you find a gin and tonic?"
Ford: "I didn't. I found a lake and through myself in and out of that."

And then, there's the joke that literally took me years to understand...

"[Going into hyperspace] is rather unpleasantly like being drunk."
"What's so unpleasant about being drunk?"
"Ask a glass of water."

When the emo Robot (marvin was his name?) beats the shit out of that hyper destruction robot with every weapon in the galaxy just by messing around by talking a lot of BS!!! ^^ laughed out Loud at that one. :)
Also a small piece of a pie (can't remember which small cake) keeps the whole universe together, damn those books have a lot of crazy imagination in them.

Not the book, or the movie, or the brilliant 80s series, but i was playing the infocom game on my phone the other day and this really made me smile. Even the game is fantastically written.

The novels are adapted from a radio tv series, and are infamous for their inconsistency between publishings. Basically, Earth blows up, and a guy in a bathrobe sees a ton of weird shit.

My favorite moment would be when I read the "original novel" in the part where the aliens are announcing to Earth that they will begin demolition, just the whole sequence made me laugh and was captured so perfectly, I think in every other medium the scene hasn't been as nicely done.
