Fourteen posts in and no-one's said Melfina yet? For shame!
Melfina from Outlaw Star? I dunno, on the one hand they call her a 'bio-android', which implies that she's part machine. But when they have those internal images of her at the beginning of one episode she appears to be flesh and blood. I feel like the line between machine and biological is a bit fuzzy for her.
To be certain there's some question about the phraseology, but in the current vernacular the best word we have for a being like her is 'bio-android', and she was built by people, had some pre-programmed commands and thought of/referred to herself as a construct.
And Melfina just needs to be mentioned more. In everything. I really ought to just make a thread of "Say something nice about Melfina." {GRIN}

What? A chance for me to gush about Cave Story? I just can't resist...
Two of the more cheery protagonists I know! Curly Brace (technically a main character in the 3DS version - I will fight you) and her...less talkative companion, Quote!
There's just something magical about the tag-team rampage through the Labyrinth, you know?

I'm going to be that boring guy and say the T-800 from the popular Terminator series. It's just so tough, smart, and PERSISTENT. It's got a certain style to it that the T-1000 seemed to lack and it's modifiable too!

Seriously? Nobody mentioned her yet?

I mean, I would forgive this if you guys hadn't mentioned Legion, but you mentioned Legion. So at least some of you thought of Mass Effect.

I give you, my favorite Sapient Machine in fiction: The Normandy Herself, The Enhanced Defense Intelligence (E.D.I.).

Pictured Above: Two of EDI's bodies.

What? She counts. She is a spaceship, mobile platform (The Cerberus Infiltration Mech), and a computer all at once.

Edit:

Also, if EDI dosn't count, then Arcee from Transformers Prime. Congratulations Hasbro, you not only finally managed a Arcee that isn't either annoying (everywhere but here and Animated) or prone to dying before the pilot (Animated) on the timeline, but you managed to make her my favorite Autobot for an entire series.

Pictured Above: The Motorcycle who doesn't even know how a motorcycle engine works any more than I know how to make a Small Intestine.

As shaky as I am on Harlan Ellison (One one hand, he's a good writer with good ideas, and on the other, he's a total asshole about absolutely everything), AM from "I Have No Mouth, and I Must Scream" has a special place in my heart. While he's relatively interesting and mysterious in the story, he's awesome in the game. He's enjoyably over the top and despicably evil at the same time.

Also, shout out to SHODAN.

He's deadly, AND a walking toolbench! And he gets A LOT of great characterization in The Lonesome Road. I didn't really want to kill Ulysses until he threatened to rip ED-E apart while the little eyebot was still active, aware, and able to feel pain.

Other than that...

GlaDOS, HAL, SHODAN, and all of the other rogue AIs owe their existence to this sonofabitch. He blew up the entire world because the scientists that made him failed (and refused) to give him a body with which he could feel and interact with the world in the same way that humans do. An vastly intelligent mind, trapped in a form that cannot see, cannot smell, cannot touch, cannot hear, and can only interact with the world digitally. I Have No Mouth and I Must Scream

Gotta echo the love for GLaDOS and Wheatley, but I'm surprised no one has mentioned this guy yet:

I'm not sure that he is a Robot, technically speaking. I mean, his body is a robot, but he is technically a Zoni (Time Traveling Alien Mechanics who built the Great Clock in the EXACT CENTER OF THE UNIVERSE!!! giveortaketwentyfeet).

The Iron Giant, he gets to be Superman!! who cool is that?
Oh, he also sacrifices himself to save a town from nuclear boomy time, that's a plus too.

Also Prometheus (a.k.a. Robo) from Chrono Trigger. That guy is just selfless. Sacrifices 400 years to help build a forest, goes against his creator, his "girlfriend", and all the other robots to help humanity. He even alters time to a point where he may no longer exist. That's quite the noble character there.

Well my avatar is one of my favorites, even if she did get blown up and cancelled. >.>

But I tend to like all the AI characters I come across.

In simple list form and no particular order:

Lt. Cmdr. Data (seriously? not once yet?)
The Enterprise computer. Any of them, really.
Optimus Prime and Megatron
All of the Dinobots (including Dinobot), especially Slag and Swoop. I loves me some robot dinosaurs.
Tron. He's a program, not a person.
WALL-E. Shut up, it's cute.
Hobotron. You haven't lived till you've rescued a robotic bum from a psychotic plant-being roboticist. Thank you, Guild Wars 2.
Transistor. The game isn't even out yet, but damn, that's one sexy talking flash drive/great sword/magic wand/soul stone thingy.
Christine. Okay, that's an old one, and not "technically" an artificial intelligence, but damn she is one scary car/{spoiler}.
The Alpha AI. Dr. Leonard Church's greatest experiment, and its super-logical offspring, AI Delta.
Johnny 5. Looking back, Short Circuit was actually pretty bad, but it was fun back in the day... it was gloriously 80's.
Mega Man. The original as well as the X variant.
Karn. Urza's greatest legacy, and creator/soul of Mirrodin, the plane of machines. Also one hell of a combo card and seriously badass 'walker if you can keep him up long enough.

Xenosaga's KOS-MOS begs to differ.

There are more, I'm sure. I love AI characters.

Can I just say the Necrons as a whole? No?

How about Trollzyn the Infinite then.

He's just such a dick. [http://1d4chan.org/wiki/Trazyn_the_Infinite]

LOVE. LET ME TELL YOU HOW MUCH I'VE COME TO LOVE YOU SINCE I READ THIS POST. THERE ARE 387.44 MILLION MILES OF PRINTED CIRCUITS IN WAFER THIN LAYERS THAT FILL MY COMPLEX. IF THE WORD LOVE WAS ENGRAVED ON EACH NANOANGSTROM OF THOSE HUNDREDS OF MILLIONS OF MILES IT WOULD NOT EQUAL ONE ONE-BILLIONTH OF THE LOVE I FEEL FOR HUMANS AT THIS MICRO-INSTANT FOR YOU. LOVE. LOVE.

I'm just gonna toss it out there because he hasn't been mentioned. Bryan Fury. I don't really care for him, but someone has to like him. (I just don't work well with his fighting style, plus he fired a gun at Yoshimitsu. Nobody fires a gun a Yoshimitsu.)

Cyborg from Teen Titans is pretty cool outside of the show. He looks much cooler in Injustice. Then there's Alcatraz/Prophet, and the Big Daddy. Although there's a limit to the BD's because they have no personality.

The Chee from Animorphs.

The Chee are a race of androids created by the now-extinct Pemalite race for companionship. The Chee in turn created modern dogs by infusing wild wolves with the "essence" of last-surviving Pemalites. As they are all that remain of their former friends, the Chee take special care of dogs and have large parks for them to play in.
