"What's the difference between you and me, kid? What makes a gifted killer like yourself into a rock-hard bastard like me? The answer is TRAINING you ignorant FUCK! I've got a hundred years of experience on you, and don't you forget it!"

John Marston: "I'm gonna hand you over to 'em and watch 'em tear you limb from limb."

MacDougal: "WHAT?"

Marston: "Just kiddin'. We're gonna run across the rooftops. Getcha' back to your ivory tower."

"Some call them demons, i call them bitches...hur hur." and
"You reap what you sow force answers force war breeds war and death only brings death. To break this vicious circle one must do more than act without thought or doubt."
Metro 2033

and Dawn of War has some nice quotes, too many to mention here, as a wise man above me said the Imperial stronghold intro speech is sweet and made me go *gulp* and shudder.
Company of Heroes has some damn funny ones, "Conrad tie your fuckin' laces!"

Any of Irenicus' lines near the start of the game in Baldur's Gate II

Your pathetic magics are useless, LET THIS END.

EDIT:
Also, "Garrryyyy...GARRRRYYYY!?" from the "Gary" Vault in Fallout 3.

My personal favourite from that is:
"I FEAR NO EVIL, FOR I AM FEAR INCARNATE!"

One of the better ones is Hitman: Blood Money in 'Death of a Showman:
'Names are for friends, so I don't need one.'
Kinda badass there, 47.
The greatest lol-worthy pre-game release speech is Johnson's in Halo 1:

"Definition: 'Love' is making a shot to the knees of a target 120 kilometers away using an Aratech sniper rifle with a tri-light scope. Statement: This definition, I am told, is subject to interpretation. Obviously, love is a matter of odds. Not many meatbags could make such a shot, and fewer would derive love from it. Yet for me, love is knowing your target, putting them in your targeting reticle, and together, achieving a singular purpose, against statistically long odds."

-HK-47

"I am Bowser, businessman of legend! Fear my accounting!"

Hold the line.

Not just that one line of dialogue but that entire speech. One of the most epic pre battle speeches in anything ever.
Meh, I always found it rather jarring how he just seems to forget what he's going to say halfway through and then the music just drops instantly.

OT:

"You know, the world isn't run by the laws written on paper. It's run by people. Some according to laws, others not; it depends on each individual how his world will be, how he makes it.

And you also need a whole lot of luck, so that somebody else doesn't make your life hell. And it ain't as simple as they tell you in grade school...

But it is good to have strong values, and to maintain them: in marriage, in crime, in war, always and everywhere.

I messed up. So did Paulie and Sam. We wanted a better life, but in the end we were a lot worse off than most other people.

You know, I think it's important to keep a balance in things. Yeah, balance, that's the right word. Cause the guy who wants too much risks losing absolutely everything. Of course, the guy who wants too little from life might not get anything at all."

So good. So very good.

[small] Yes, I'm aware that that's more than one line.[/small]

I have a couple that spring to mind.

"I, Garland, will KNOCK YOU ALL DOWN!" - Garland, FF1. It's a classic that makes me laugh every time.

"Despair, so delicious; Fear, so exhilarating" - Marwyn(Or Falric, I can never remember), WoW. The voice acting for this line was pretty sweet.

"Knowledge is power. Hide it well," - Warhammer 40,000: Dawn of War

Additionally, StarCraft has a few awesome quotes

"You speak of knowledge, Judicator? You speak of experience? I have journeyed through the darkness between the most distant stars. I have beheld the births of negative-suns and borne witness to the entropy of entire realities... Unto my experience, Aldaris, all that you've built here on Aiur is but a fleeting dream. A dream from which your precious Conclave shall awaken, finding themselves drowned in a greater nightmare." - Zeratul

"It is difficult to believe that I am working along side my greatest enemy in order to save this sector. Fortune has been whimsical of late." - Fenix

Kerrigan: "Now, are you ready to die a second time?"
Fenix: "The Khala awaits me, Kerrigan. And although I am prepared to face my destiny, you'll not find me easy prey!"
Kerrigan: "Then that shall be your epitaph."

- All of Duke Nukem's famous out-of-context action movie quotes (especially what he says to the Overlord before he fights him, and what happens after he wins, in Duke Nukem 3D.)
- Anything and everything said by SHODAN but especially "You murdered their young!" (how can that NOT be disturbing?)
- Ditto GLaDoS ("Any contact with the chamber floor will result in an unsatisfactory mark on your official testing record, followed by death.")
And of course the old classic: "Sheep have many uses." Just for pure "WTUF?" value.
