The beginning of Bioshock is one of, if not the, absolute best introductions of any game this generation. It does an excellent job of setting up the games' tone and mood and establishes its setting, story, and some of the themes it's going to explore right in the first ten or fifteen minutes.

Sorry, I'm replaying through it now and failing to remember exactly why I never claimed it was a great game. Maybe when I get to that fucking camera or do the umpteenth fucking fetch quest I'll remember, but until then I'll blissfully ignore my previous thoughts and roll my eyes at the damned hacking minigame.
Quoted for truth.

Also, I'm replaying the game right now, and I have to say, it's MUCH shorter if you use the New Game+ feature. Since Plasmids, weapons, and items carry over, you can skip a surprising amount of the side quests. For example, Medical becomes a pretty straight shot to the boss when you don't have to go get Telekinesis, and if you collected enough components in your first run, you can skip the entire Farmer's Market section when you're making the Lazarus Vector.

Plus, experience from photos carries over too, so you don't have to do that anymore, either. And, since you'll be basically tripping over money (since you'll have so much ammo to start with), you can just buyout any hack instead of the minigame.

The Nalorakk encounter in Zul Aman on WoW. He says in a psuedo-Jamaican accent, ''You call on da beast, you gonna get more than ya bargained for'' Always makes me smile.

Riding into Mexico in Red Dead Redemption with "Far Away" playing in the background, and the sunset just peeking over the canyons. It really was magical.

The levels Ghosts and Anomaly in Metro 2033 where Artyum and Kahn walk through the tunnels and he explains his philosophy.

Red Dead Redemption, after you finish "And the Truth Shall Set You Free", when you're riding out of Tall Trees to Breecher's Hope and they play that song, I think it's called Compass, that was a hell of a moment.

No one mentioned Dead Space 2 yet?
Say what you want about that game, but it most certainly had some awesome gameplay sequences...
I really need to finish that game.

OT:
-The train level in Uncharted 2. Nothing in the sequel really topped that in my opinion.
-The battle against Magus in Chrono Trigger.
-The ladder from MGS3.

Cid's speech in FFVII was a pretty good one. For someone who acted as comic relief the whole game, he knows how to motivate people. Also, when everyone shows up on the Highwind to go finish the game was a pretty cool moment.

I've never really felt for characters in any other games besides RPGs because I'm usually playing with silent protagonists, but there's probably a few moments I don't remember.

Uncharted 2 had plenty, but my over-all favorite would have to be the heli-copter chase scene on the train.

Pripyat in any of the STALKER games is amazing, just wandering down the empty overgrown streets dodging the packs of feral mutants and hostile mercenary is a fantastic experience. Exploring the old abandoned (And sometimes only seemingly abandoned) buildings in search of elusive artifacts and stash never gets old for me.

There's also a scene in Neverwinter Nights 2: Mask of the Betrayer where...

You're standing atop the massive skeletal corpse of a dead god as it floats through 'the nether'.

which I consider bloody brilliant. One of my favourite moments in a fantasy rpg right up there with challenging the Bhallspawn in Baldur's Gate 2 and confronting the 'villain' in Planescape: Torment.

Well, I have a few:

1) Chasing the Black Dahlia Killer through the tunnels in L.A. Noire
2) Driving the snowmobile in Modern Warfare 2
3) The last mission in Mass Effect 1

And finally...
4) Blowing everything in Venezuela to hell in Mercenaries 2 while listening to an assortment of epic music on my iPod.

Well I just finished Uncharted 3 so I might be a bit biased...

But the scene where you're in a castle in France, and then things get a little, erm, hot.

It was so beautiful.

Pretty much all the set pieces from that game had me practically drooling into my controller.

I always loved the Fallout intros as well. Super atmospeheric.

Wind Waker on returning to the forsaken fortress, you come back geared up ready to take on enemies as you slash your way through your previously unbeatable enemies, you reach the tower save your sister, run from the helmrock king (massive bird) that is destroying the spiral path up the tower as it floods with water and you reach the top for an awesome fight with intense music and you smash it's skull with a hammer and then you face Ganon...dorf! that as a 10 year old kid just blew my mind

Ending of Halo: Reach. We all knew how it was going to go, and you went out like a badass to the end.

It's like the developers understood how frustrating the rest of the combat was in the game, so right before the final boss they gave you your revenge, in the form of an axe weilding maniac with a ridiculous amount of health.

I remember first starting out in WoW, back in Vanilla, Only went to lvl 60 times, everything had been pretty much standard fantasy fare for starting out as a Night Elf. Then in Darkshore, I came over a rise and came face to eyehole with a massive, alien skull. That's burned in there pretty good.

Best sequence ever though? Okami. The Final Battle. The cutscene with "ReSet ~ Thank you Version ~" and then into "The Sun Rises" as you unleash an industrial sized can of whoop-ass. I was crying Manly Tears through that whole battle.

I think it might have been nighttime for be, which made it all the lonelier. Great great gaming moment, either way.

OT: I know this is cheating, but the entire middle part of Uncharted 2, from when you board the train to the end of the Himalayan temple sequence was peerless video gaming for me. One epic jaw drop after another. It made the final thrid of the game seem pretty uninspired by comparison.

The assault on the penitentiary in Secret Armory of General Knoxx. Mini Steve and Mister Shank!

The final segment of the Jungle section in Killzone 3, with the attack mechs and dozens of Higs swarming you as the smoke drifts in.
