Well... I found the cast of the Tak trilogy to be pretty funny... I think it's mostly due to comedic timing during most of the character interactions...

I would say the same thing for the cast of the Hyperdimension Neptunia series... but I think it's mostly due to what each of the main characters were based on which sold the genuine humor, in my opinion...

Sticking with the Mass Effect trilogy, I find Wrex to be really funny and naturally so - he's not playing for laughs, it's his manner and way of speaking that raise the chuckles, especially in ME3. Joker does have some zingers too, I will give you that!

Most of the ones I'd bring up have already been said, but I'm surprised that no one's mentioned Fidget yet. The delivery of every single line is absolutely perfect and just makes her dialogue hysterical.
"You know what? I kinda hate this guy, and I would love to maul his face off."

I think Portal 1 + 2 are literally the only games where I found characters to be consistently funny.

Tiny Tina.
She's great. Between using slang you'd expect from a rapper to delivering some of the best lines of dialogue in the game. She's by far my favorite character of the series.

Jack from Vampire: The Masquerade Bloodlines.

He straddles the wonderful line between funny and badass.

Also officer Chuck from the same game... that loveable fat useless bastard.

The interactions between everyone in the investigation group in Persona 4 is pretty much always rather funny, so I'll go with that.

The Black Whirlwind from Jade Empire. All interactions and dialogues with him were great. I loved a brief conversation he had with Kang the Mad toward the end:

BW: Kang, can you invent me a bowl I can use to drink wine with stumps?
KtM: Yours or somebody else's?

And the bit where you play him against the paratroopers and the Goldem, with the UT style commentator. Pure class.

Also, HK-47.

Persona 4's Yu Narukami. The anime basically uses all of his funny choices, IIRC.

Oghren from Dragon Age. The interactions between him and Alistair are chuckle-worthy.

And of course, everyone's favorite Merc With A Mouth, Deadpool. The Wolverine slapping scene was way too much for me.

There's a heap of funny moments that I love in the Mass Effect series - if I had to pick one character though it'd probably be Mordin. The G&S song, "might run tests on the seashells", his little talk about human-turian relations, so much gold.

I enjoyed Shale a lot in Dragon Age: Origins. HK-47 was fantatic as well, especially in KOTOR II where there was a bit of subtlety to balance out all the meatbag one-liners.

Going back a ways there were a lot of classics from the old Lucasarts days like Sam and Max and Guybrush Threepwood who were great.

Agent York's mental instability in Deadly Premonition has a special place in my heart.

Also, the cast of Psychonauts. The asylum residents and camp councilors in particular.

Just about everyone among the Dragon Age and Mass Effect crews had some good banter going for them, but Mordin's style of humor stuck out for me as well. An eccentric genius done right.

Not sure if funny is what I like about game characters but I do like psychotic game characters who aren't over the top with it. Like if you were in real life and you met this person you'd think they were almost on the edge of shanking you and urinating in your eye socket but they haven't broke the line yet to where they seem like an actual threat. Kind sounds weird when I read it but yeah lol.
