Just Cause 2: Finding a random hot air ballon in a valley. Which you can even use! And this was after I'd clocked over 100 hours on the game and thought I knew everything about it.
Same bro, when I found the hot air ballon, I spent like 15 minutes figuring out how to use it. Then I found all you had to do was shoot off the sandbags. FML. Yeah, I have platinumed Just Cause 2.

Any of the Colossus in Shadow of the Colossus. In Bad Company 2, anytime that my team had organization, like the Helicopters were in sync, as well as the tanks and the infactry, just like you see in the movies, those are the greatest moments ever in shooters in my opinion. And Kingdom Hearts 2, when you fought the field full of enemies, and when I beat Sephoroth for the first time.
I so agree with the Bad Company 2 bit! It is AMAZING when tanks, infantry, and air support are all used in chorus with a great team of people.

Just 10 minutes ago I left BC2 after a great sesh with my mates. Somehow, I managed to go up learly 2 levels today! (26-27.5)

Half life 2 episode 2 when you're armed with a turret, two resistance fighters and a cache of infinte shotgun ammo. Strategically splitting our forces as the waves of antlions got progressively harder then realising the turret has failed, the two resistance fighters wet their pants at the sight of a maximum danger level from all entrances.

Silence...

A rain of corpses follow as the antlions fall dead from their holes.
The vortigaunts had arrived. The following firefight with one of the vortigaunts and me at the front protecting the rest from antlions which sailed around the room felt biblical. After the carnage I simply had to go back and look at what I'd done. "Holy f**k that was awesome!"
Easily my favourite set piece in any videogame ever. I cant wait to see what valve does to top it for Episode 3.

Saints Row 2 just about all of it. DE:HR two guys were standing next to a fire extinguisher and I shot it with my silenced 10MM then as they were confused I did a double take down. The air field raid in Medal of Honor(2010). And even though I hated the MP of Halo: Reach the SP was good and the ending seem was epic!

There's been a few over the years but some standouts:

Mass Effect: The opening drew me in immediately and the ending left me in a state of pure satisfaction. The ending felt complete (no stupid cliffhangers to make you buy the next game) and I was looking forward to the next chapter.

ME2: More awesomeness like others have said.

First time I played Planetside. I couldn't comprehend the awesomeness on my screen. Dozens of players trying to hold off an epic assault with tanks and gunships about. I think I was just running around like a headless chicken firing wildly towards the enemy. The grin on my face from that experience is unforgettable.

The first Alien vs Predator: I crapped myself playing as the colonial marine. Alone... super dark... super fast aliens... that bloody motion tracker sound that still haunts me every time I hear it.

Homeworld. The whole game was one amazing experience for me and it worked on so many levels. Ahead of its time, I loved the story and how it was tragic without having to rub it into your face and remind you every few seconds. You knew what happened and that you had to give it your all to actually make it to the end of the game. Lose a unit? You wont have it for the next mission! Also, the use of excellent music, including Adagio for Strings:

There's plenty others too.

Crysis - heading downriver in a boat, only to find myself tailed by a helicopter. I hide under a bridge just in time to dodge the chopper's missiles, pull out my rocket launcher, and bring it down with two direct hits - at which point the flaming chassis falls from the sky straight towards me, skidding to a halt less than three feet away from where I stand.

All in all, it felt like a scripted-cutscene action-movie moment.

The most recent one that comes to mind for me is playing the demo for Space marine, the first encounter with orks was a holy fuck this game is awesome moment.
Same for the short jetpack section you can play to. Games need more jetpacks.

Call of Duty 4's All Ghillied Up just.. had me. I cannot explain it any further.

The other Vaults, bat shit randomness and epic moments in Fallout 3, no other game like it. 'Give me the naughty nightwear'

Getting 'wasted' in GTA IV by the most extravagant ways was awesome, best was when I survived jumping out of a helicopter basically nose-diving.
Lot of other GTA moments, I simply cannot fit it into one post. What a series.

For MGS, refer to above. Final battle of MGS4 was immense.

So many others, even in sport games where you score a insane goal in extra time from outside the box. A clutch 3 Point shot, etc etc.

The first time I juggled a staggered enemy in FF13 was pretty damn epic. Hell, the first time I juggled a BOSS was better.

Exploring the Mile High Club in Just Cause 2 was good, too. And then taking the jet out of the back. :)

All Ghillied Up, from CoD4. Amazing mission, really made me feel like a badass, unlike MW2, where it just felt like all you were doing was watch the awesome moments, instead of actually playing them.

The first time I played Dead Space 2, that intro got me going. It was 1 in the morning, surround sound blasting. I played through most of the game that morning.
