Mass Effect 2 was pretty awesome throughout, but the part that stands out most to me is when
the Collectors board the Normandy and you control Joker as your crew gets captured.
That part was just so well done.

Spec Ops The Line: white phosphorous.
Ninjas. Damn.
By that part in the campain I was ready for anything. And then the game decided "Nope. Time to crank it up to eleven."

For me it would have to be "Three Leaf Clover" in Grand Theft Auto 4. It's easily my favorite mission in that game, even if it is fairly easy, it's just such a thrill to run down the streets of Liberty City lugging around a million dollars in duffel bags with the fuzz descending upon you.

Assassins Creed 2

{When Ezio Finds his family strung up on the gallows, betrayed by his father's trusted friend}

That, combined with the rest of then intro really pushed me over the edge for the rest of the game

The first Grandia game, when you climb the huge wall at "the end of the world" reach the top and see the rest of the world beyond it.

You haven't reached the end of the world, on the contrary, the adventure is only just beginning.

Before you lies a huge new strange and wonderful world and its going to be so much fun to explore it all!

System Shock 2. SHODAN reveal. Holy Jesus.
Oh yes. Definitely saw it coming (really Polito? Nano-chambers are preferable to slow decomposition? Your OS is showing) but it's a great reveal.

That was the turning point of the game for me. It was like I was just getting the swing of things, my character had become fairly beefy, and I had figured out the little "dances" you had to make with each enemy to take minimal damage. Then you get to Deck 4, the monkeys can kill you in two hits, people are being re-assembled into ninja-cyborgs, and the grunts get goddamn BOMBS. I had to stop and listen every time I found a new room because I was terrified of what lay ahead. And sometimes failed to notice what was stepping up behind.

And I won't spoil anything, but people who have played the game should know what I'm talking about when I say the Body of the Many - the way you enter that level, and the total insanity of what follows, is an astounding exercise in atmosphere creation. And what happens after that is even better.

KIngdom Hearts 2, when Sora first gained the Drive gauge and I saw him dual wielding Keyblades... I couldn't stop gibbering like an Overlord minion.

Dark Souls, going through Ash Lake and finding the Everlasting Dragon.

The Would you Kindly? moment.

Meeting Vaas in Far Cry 3, yeah it was at the beginning, but damn.

Half the shit in the Mass Effect series as well

Mortal Kombat, fatalities. It was hard enough just trying to catch a glimpse of the game at the arcade (I was much shorter back then), but with the words finish him the view became much more obstructed. The game was already awesome, punching off heads made it amazing.

Man I miss Grandia...

OT: Pretty much the whole Mass Effect trilogy was full of those moments.

Mass Effect 3, when the battle for Earth begins...Gawd that was epic
Hell yeah, Mass Effect 3 has had a lot of flack for many many reasons but the whole assault on Earth rocked my socks off. I really should replay this game, probably when they stop releasing DLC for it.

On a personal note, when I first started playing Uncharted 2, I was pretty unimpressed, it was alright but not the thrill ride that everyone promised. Then I got that bit in Nepal where the helicoptor attacks you and you have to escape through collapsing buildings and what have you, holy crap I was hooked after that.
