Definitely the StA5X Arc Cannon from Killzone 3. You would just pull the trigger, watch the green orb home onto an enemy and BOOM! Good luck finding him. It was so satisfying doing it over and over and still I replay that one level just to use that beast.

I would say that it has to be the Gravity Gun in Half-Life 2.
Special mentions go to the portal gun, which isn't a direct weapon but could be used to inflict some nasty damage. Also, the fusion rocket launcher in Red Faction. That thing would kill you if you were anywhere near the object you hit... it was sooo massive that the whole right side of the screen was obstructed while you armed it.

Oh hell yes with BioShock, when you get Insect Swarm fully upgraded you can drop splicers three at a time.

The chaingun eats flesh like a starving pig, the Lion Heart looks the part and deals a fantastic limit break finishing blow of the same name, and the RCP-90 is simply death incarnate and my favorite way to wind down after a hard day at the office.

In my own opinion, it has to be this, but only in Call of Duty 4; I have no idea why I like it in the aforementioned game, possibly because it feels so out of place (what, being 60 years old and all), but I like the sound it makes and it works for me;

The Morph Gun from Jak 3. 12 forms, including a nuke, if thats not awesome then what the hell is?
The lancer from Gears of War. It's a large machine gun with a chainsaw as a bajonet, that's it. It's simple but effective and gives a satisfying feeling when you grind it through an enemy's digestive system in alphabetic order.
PS: I would like to mention the SPARTAN laser because in Halo Reach it's powerfull enough to destroy or unarm a tank in one shot. Unfortunately it takes time to charge it's laser ( SHOOP DA WHOOP!).

I'll probably think of a better one later, but the one that first popped into my head was the Railway Rifle from Fallout 3. Far from the most powerful weapon in the game, not particularly flashy or useful, but I just never get sick of seeing a railway spike rip off someone's head and nail it to the wall in slow motion. Immensely satisfying.
I was just thinking about this a few days ago, actually, and it's definitely ChaosUT's Gravity Vortex for UT2004.
