Mario Kart DS - Great racing game to play alone or with some friends.
Metroid Prime Hunters - Great FPS Adventure, really makes good use of the touchscreen.
Resident Evil Deadly Silence - Nice port of the original Resident Evil with some extra stuff added.
Grand Theft Auto Chinatown Wars - Nice little sandbox game. Great for a portable GTA fix.
Diddy Kong Racing DS - Great rerelease of the N64 classic.
Super Mario 64 DS - Another N64 classic.
Sonic Rush - As a longtime Sonic fan, this game miraculously made it to one of my favourites of the series. Total must buy for Ds owners.
Chrono Trigger - Great rerelease of the SNES classic. Widely known as one of the best JRPGs of all time.
New Super Mario Bros - Brilliant Mario side scroller similar to the classics.

I suppose Nintendogs. They're good sim games, and they do make good use of the DS features.

Any other Mario game, the RPG's are surprisingly good.
Pokemon is always fun. HG/SS just released...

Super Robot Taisen OG Saga: Endless frontier, an excellent JRPG with a funny cast, great battle system, and it will last you a while.
Tetris DS, its Tetris....need i say more?
Kingdom Hearts: 358/2 days, its amazing and will probably make you cry
Legend of Zelda:Spirit Tracks
Mario&Luigi:Bowsers Inside Story
Fire Emblem:Shadow Dragon
Bleach
ark Souls, a great 2d fighter, it can be played multiplayer even if only you have the Game
Bleach:3rd Phantom, a nice JRPG that's like a mix of FF Ta tics and Fire Emblem
