Q: Do You think you could take a life to defend your own?
As I've never been in a situation where my life has been threatened in such a way, I cannot give an absolutely certain answer as to how I would react. However I do think that yes, I could take a life in self-defense if it were necessary. I wouldn't like it, and if it were reasonably possible I would attempt to disable them rather than outright kill them (though I wouldn't feel guilty if I did kill them, and I wouldn't increase the risk to myself just to ensure a non-fatal elimination.
Q: The BadGuy is threatening not just your life, but your family's. Does this alter your reaction?
It does not.
Q: Would the difference between the up-close kill, and the point and click of a ranged weapon have a difference on you?
Aside from the obvious increase in difficulty and physical effort a hand-to-hand fight would provide when compared to just pulling a trigger, no. There would be no difference for me morally or pyschologically.
