Fear Effect had two gay women as the leads. For most games it wouldn't matter because it would never come up. Is Samus straight? She never has contact with other people in games so it never comes up. Almost all romance in games is ham handed and badly carried out so it doesn't really matter if they are gay or not, it usually doesn't come into play.
I can't tell if you used the example to make your point or if you are honestly asking. If it's the former, consider this expounding on the point. If it's the latter, the answer to your question:
Samus might be an attractive young female, but she's also got a super dangerous career (and life's mission, if you believe certain tie-in materials) and an enemy list as long as her arm who would be quick to kill someone she loves just to make her suffer. I can easily see where she would have neither the time nor the desire for romance in her life. (No, Adam from Fusion doesn't count.)
In a perfect world, romance, be it gay, straight, or any other orientation in the world, would be handled realistically and gracefully, without the hamhanded "we're just tacking on the romance because it sells" feeling. I am a fan of a good, solid romantic story, no matter what the sex of either party involved is. But personally, I wouldn't worry about a gay romance plot until they can perfect a straight romance without making it feel superficial or stupid (and yes, I am well aware there are good games out there with great romance plots. I just don't believe they're in the majority).
For the record, I voted "yes", because I played it as "okay, let's say it's a perfect world, where people can write a good romance where the characters are well-developed as individuals, and their romance is realistic in its development and yet doesn't interfere with the other aspects of the story in any significant way. If they can pull that off, then I think people wouldn't or shouldn't care if both parties are male or female".
