I'll be using the Commando first, partially because the support gunner class in Borderlands 1 effectively removed ammo from the game while increasing my damage and mag size and partially because the Gunzerker/Siren look wierd and zer0 doesn't fit my playstyle.
Although once the Mechromancer comes out, I'll try her because she seems quite interesting and having a giant robot(?) on my side is always a plus.

Me and some friends enjoyed the hell out of the last one. I ended up being Berserker so I imagine I'll land on gunzerker for group play and mechromancer for solo.
ill start with the commando, but i will more than likely play the mechromancer. I am a sucker for mechs, plus her art design is cool.

I was always a Siren in the original, so I see no reason to stop that tradition. Plus, she has healing bullets. That's fucking awesome.

Axton, if he still has Roland's trademarks (ammo/health regen on demand, and sniper-shotgun mastery). If not, then I'll play a little of Maya and Salvador and go with the one I like the most.
Axton looks like Roland done right. Ambush power combined with turrets to draw away enemies. Looks amazing.
However, I'm curious as to see how the Mechromancer's ability works out. Will it be just another ability to summon an NPC companion? Or will gearbox devise something more... devious? Like a giant robot you can assume direct control of like a drone?

I started up a new game of Borderlands in preparation for BL2 lol. I picked Lilith, again. I tried Mord 2 times, and both times I just couldn't stand it. I tried Brick once, then realized that since I don't use rockets and his fists kinda suck he has nothing to offer me. I tried Roland once. When I saw that I hated his skill trees, mostly because I don't use CRs and I prefer shotties with LESS spread, I promptly ended my soldiering career.
Lilith just has too many good skills. Even if you took phasewalking completely out of the equation I would still pick her. Also she has the only voice/lines I actually like.
The thing that I'm loving about BL2 is that I am honestly having a tough time choosing who to play as. Well I'm not playing as Salvadore, that is a given. I appreciate this playstyle and skill tree. I think its pretty cool sounding, but its not for me.
I'm leaning heavily towards Maya, much like always playing Breton in ES games, and Wanderer in Souls games, I have a history with Sirens.
I also use primarily elemental weapons, and cool lasers whenever possible. Sniper, SMG, Shotgun, Revolver are my 4 go tos.
Axton's skilltrees are amazing, and I can definately see all kinds of ways to build him. As long as he doesn't have some douchey "bro" voice I may use him.
Zero is basically a lot like Lilith. Going stealth and popping out for major mellee damage, combined with sniping, what's not to love?
I don't really like the look of Zero, or the voice I've heard. Also the way the screen darkens and the enemies weakpoints are highlighted in stealth is really distracting. I'm sure its helpful, but it just seems like I'll be seeing that visual chaos a lot if I use him so I had better like it or pass him by.
Gaige doesn't have more than 2 known skills so far, but since I play a class on an MMO that uses summoned creatures a lot it will probably feel very familiar.
The only things I don't like about Maya are her look, which can easily be changed, and the Phaselock seems kinda unimpressive compared to the other action abilities. I haven't seen her gamechangers to it in action yet though.
As long as their voices don't suck I guess I'll play Maya or Axton first, then Gaige.
An option to disable your characters one liners completely would be great.

Zer0, seems to be built similarly to the Mordecai from the last game or at least seems that way. I'm sure there are some big differences but it will be the first char.
I want to play as the mecromancer... so it looks like I will be a siren again until that 2-3 months when I can play as an awesome bot-weilding killer babe!
I'll probably wait until I know more about how they play before I commit to one for an entire playthrough, but going off looks alone I'd have to pick Zer0. As cool as the others (especially Maya) look, there's just something awesome about him.
I'm terrible when it comes to shooters, so odds are good I'll play as the Commando so I can have some support fire. Still need to see how the Mechromancer plays, but might go with that on a second playthrough.
