The Void, I know it came it out last year, but that game deserves to be game of the year. It's the only game that I can call high art with a straight face, and for that it deserves to win more awards.
No love for SSF4?

So far its been Red Dead Redemption to me.. Truly stunning game..

I have both Civ V and New Vegas from those. It's a very hard pick between those two as I love them both and haven't really had the time to play either enough yet to get really into them. I'll go with Civilization V as I've waited that for longer.
Civ V.
1) It's the only one (out of the list) I've played
2) It's awesome. (I still find myself playing Civ IV though... I guess that was just slightly awesomerererer in my opinion...and because you can't be Stalin in Civ V)

OT: Mass Effect 2. The only other games that I feel are GOTY-worthy are RDR and SC2. And Gran Turismo 5 isn't out yet, but if it comes out in 2010 it will be a definite contender.

Transformers: War for Cybertron. Although, I expect Fable 3 to contend that, but I cannot say for sure it will take the title because alas, I havn't played it yet.
Also, OP, your poll options make me feel like my opinion doesn't matter, because I do not consider any of the games listed in the poll to be my favourite release of 2010. No offense, just sayin'. (my opinion doesn't matter anyway

Mass Effect 2, obviously.
I've never been so pulled in by a game franchise before. We're all on the edge of our seats waiting to complete our personal stories with the trilogy finale.
This has spanned years, mind you. No other game, that I can personally remember, has been so breathtaking. They really pushed the envelope with ME2. The only people that didn't like Mass Effect 2 (or 1) that *I* have seen, are people who say.. "There is too much talking." It's an epic space-opera folks.
Red Dead Redemption would be my close second. However, it didn't really innovate in many departments, other than stunning atmosphere... aand I'm not exactly still on the edge of my seat from anything in Red Dead Redemption.
