And a question about Gears of War 2, what was the point of going to that creepy lab where you confront the sires? Was it a lab that tried to make humans into locusts?
The splicers originally used plasmids just like you, but when they became addicted they ended up taking it from anywhere they could get it (including straight from Little Sisters, hence, the Big Daddies) the ADAM would have been so mixed up that it probably wouldn't have any single affect like the main characters, although some do retain specific powers.
The Vita Chamber in Ryan's office was turned off, presumably because Ryan had given up and didn't want to come back simply to be killed again. There was/is a lot of speculation as to whether or not this meant that he came back alive in a more distant one, but nothing confirmed.
As for Gears of War, I believe the Sires are linked to the humans
and the Locust, again this is cause for a lot of speculation but nothing has been confirmed. Some believe that the Locust were once human, others believe that they were created
by humans. Niles states that the Sires are the "genetic link to our future" which suggests that they were attempting to create a hybrid species or evolve humanity, possibly to end the war with the Locust.

As for the Vita chambers its a plot hole just to make gameplay feasible. If NPCs could use them then splicers would just keep respawning.
Not quite. It is mentioned in the audio-diaries that only people with Ryan's genetic code can use them, as the player is his son that means he can.

Regarding Andrew Ryan's death, I haven't played the game (want to though), but I've seen that scene on Youtube and from what I can gather, he made no efforts to resist Jack killing him. He could have easily just said "Would you kindly not kill me?" if he didn't want to die, so I got the imppression that for some reason he actually wanted Jack to kill him. Correct me there if I'm wrong and have completely misread that scene.
Pretty much yes. He had the option to order Jack not to kill him, but chose not to. As to why, we can only speculate.
