While he does have a decent amount of viewers it really feels like this guy should be rolling in viewers considering how funny his stuff is.

While very niche to fans of League of Legends, still really funny.

Mathewmatosis:

http://www.youtube.com/channel/UCb_sF2m3-2azOqeNEdMwQPw

He does some of the most thorough and in depth reviews on youtube. He basically breaks down each component in a game from beginning to end and focuses solely on in-depth analyzation and insightful observations of a game, instead of diverging into unfunny jokes for about 3 minutes like most Jontron clones video game reviewers on Youtube do. He's done reviews for every single MGS game and 3D Legend of Zelda game. I recommend starting with his ICO review since it's the shortest and will give you a good idea of what his reviewing style is like.
I see your Mathewmatosis and raise an Errant Signal.

While he's not as in depth as Mathew in truth he really doesn't need to be. To me he's a little bit more sincere in his critiques and more concise, all the while still being intellectually stimulating. He also does a little bit more than review games, he sometimes talks about gaming as a medium in general.

I would also recommend the Loading Bar but Spill has shut down since late last year sad stuff. They were very entertaining.

If you like movie reviews with an emphases on the industry I recommend Beyondthetrailer

Doug "TestZero" Hancox of DragonsLayerProd.

He hasn't put out much recently, but he has two main series:

"Video Game Bunker" is yet another nostalgic video game review show. Except the framing device is that it's after an apocalypse caused by escalation of the console war (seriously). The jokes can range from damn funny to totally unfunny, and Zero's costume is kind of terrible, but he has some pretty good analysis of the games he looks at, and his often controversial opinions are worth seeing if nothing else.

"Commercial Failure" is kind of like Mike J's "Infomercialism," except that it predates it. It's also pretty funny most of the time, and some of these products have to be seen to be believed.

There are other things too, like "First Impressions Friday," "4up" (four-player speedrun contests), and the podcast "Gamers Without Borders."

Vlogbrothers always get more recognition since they actually do something to help the world. They won't rock your socks off with video games and shit (Although, they do have a gaming channel). However, they will give some decent advice once in a while.

I really enjoy lets plays from Griffinlobster and Odinspack33, especially when they get together and either guest in each others video's, or play together. They make a very good double team, but they are very good separately as well. I am rather enjoying Griffinlobster's Rayman legends lets play at the moment, and his Psychonauts let's play was also very entertaining and quite informative.

This guy has some superb music on his channel. Microsoft really should have just paid to have his cover of the Killer Instinct theme in their new game. http://www.youtube.com/channel/UC1Svb88nggK2YNrffJOSBEQ

Also Good Bad Flicks, a movie critic who will actually defend the likes of 'Prophecy' and 'Spice World The Movie' not because their ironically hilarious, but because their genuinely great fun! http://www.youtube.com/channel/UCtKttDTGqEbBrC7jDN2_axQ

Well I have two, the original I was going to say, and then the one more closer to the really obscure sub count of 1000.

My original would be brutalmoose http://www.youtube.com/user/brutalmoose . He reviews video games in the same sense that others like JonTron and ProJared do, but he doesn't have frequent updates nor does he do the latest game releases, typically staying to older, kid friendly games. One of the things I love about him is that he isn't one of those yelling/ranting reviewers that youtube has, and the kind of jokes he makes in his reviews are the kind of jokes I feel like I would make if I were doing the same thing he was, so I guess that's a big reason I like him so much, that and he seems like a really nice guy.

This next one is slightly over the 1000 mark, though only by 796 subscribers (1,796 subscribers). Called TheAnnotatXperiment http://www.youtube.com/user/TheAnnotatXperiment , its basically a channel where a group of random people have access to the channel and basically do an MST3K/Rifftrax of old cartoons and educational shorts. NSFW for the most part, and there's what I would call "4chan" humor in some of the annotations, like jokes and jabs you would expect to see on 4chan, but it isn't a constant thing, just on occasion certain things will stand out. One other thing is that there tends to be a lot of annotations that pop up at times so you'll have to pause the video to read what people wrote, which drags out the length of time on videos. Updated pretty frequently, from what I've heard they are just doing it to entertain themselves and don't really care about subscriptions. How funny they are fluctuates, but I usually get a chuckle or two out of most videos they post.

Rage Select. Jason and Jeff are hilarious, and they update daily. I've watched their naterial for years now, and it never gets old. Their reviews are as sensible as they are funny.

Look into the madness:

And their other posters:

Game Grumps deserve all the subs. They're amazing. And shall continue to be. Forever and ever. Amen.

Praise be to the Barry.
Holy be his name.
Amen.

But seriously...
There are very few to no channels that compete with the Grumps' uber-consistent episode release schedule.
Their rise on YouTube is nothing less than meteoric. In less than a year and a half they've risen to become YouTube channel #378.

And unlike PewDiePie, the king of YouTube, the Game Grumps have attained that success purely on the grounds of being hilarious and awesome.
They're hot on the heels of TotalBiscuit who has been on YouTube 6 years longer than them.

So, while one can say that small YouTube channels might be great and deserve more recognition than they currently get, one can also say that a channel as massively successful as Game Grumps has firmly established itself as a masterpiece of YouTube, and that the people who have yet to start following them are roughly equivalent to people who've failed to watch Game Of Thrones or Star Wars.
Game Grump EASILY deserves to overthrow PewDiePie from the throne of YouTube, meaning they deserve the viewership to make up that difference.

Are they the best channel on YouTube? It's hard to be sure, but it sure seems like they're at least one of them. Until their YouTube ranking more accurately reflects that, yes, they "deserve" more recognition.

-------

On a less Game Grumps-y note:

iSometimesWriteMusic deserves a TON of attention. He creates probably the absolute BEST covers of game music (and other music) I've heard.

OminousVoice, likewise, is fantastic in the music covers department.

Welcome To Night Vale deserves a lot more attention. It's getting more popular, but it certainly deserves a bigger audience. It's incredibly unique.

DNews needs more viewers. They belong to Discovery and put out tons of great and interesting science news videos every day. The more people watching DNews, the less retarded our world will be.

Shots Of Awe - Jason Silva is brilliant. He can focus the majesty of the universe, life, and reality into concentrated bursts of thought. The man is a true thoughtsmith, if ever there was such a thing. His channel needs LOADS more people paying attention to it.

--------

I don't really have any tiny channels on my radar that are good enough to say they deserve special attention.
*shrug*

EDIT:
I tentatively second Markiplier. I'm familiar with him as a guest on all manner of things I watch, though I've never been to his actual channel.
He seems pretty great, and he's on the list of channels I need to investigate.

He is the only person I can think of at the moment without going though all my subscriptions who I believe deserves more attention. Everyone else I either can't think of or has been completely inactive for several months, or a year, or more.

I like Blind LPs and Races. Lately, Lucahjin and Jay Lantis formerly Lantis02 are my favourites for that. Probably the cleanest and most updated 4-player races around.

If you've even got a passing interest in Transformers, toys, or robots in general, then I heartily suggest the always-fantastic Thew, of Thew's Awesome Transformer Reviews:

He's probably my favorite toy reviewer on the internet. Better than TJOmega, better than Optibotimus, and right up there with Vangelus and Peaugh in terms of how good he is. Seriously a dude in need of more recognition.

I very much endorse Thew.
Thew is awesome.
You don't even really need to know about Transformers.
His ridiculous enthusiasm is hilarious and great.

He's got funny material, along with a really funny series called "My Sith Is A Jerk"(although he hasn't released a new video for the series in a while).

He also made this little gem.
Currently he only has just over 600 subscribers, which is a shame because a lot of his videos are great and deserve more views than they get.

Both these guys are pretty fun to watch. Quill has some incredible stuff for strategy games like EU, CK and Civ, and he explains why he's doing things and how things work and is fun to listen to - he also has a few guides, and does some live stream chats while he's programming. Hypno is also quite fun to watch, perhaps not the most knowledgeable of finer details in games and he does miss things, but he has a good personality and his videos are generally pretty good.
