-The final boss in Jamestown ... took my like 20 tries to beat ... solo
-Jinpachi Mishima and Azazel from Tekken
-Seth from Street Fighter IV
-Prince of Persia had a couple of these ... I think the most frustrating one was the twins from Two Thrones
-Eater of Worlds from Terraria ... but whenever he kills me he dies a few seconds afterwards so it's not so bad
-Diablo from Diablo II ... mostly because I was scared shitless ... also Duriel

If it makes you feel any better: it slowly gets easier after that one. I beat him the first time by spamming ebony and ivory from across the room for like 30 minutes.
Protip: if you pauze and quit the mission right before you die, you keep the red orbs you found up to then. That way you can at least buy upgrades so your chances improve a little every time.

the big orange dialga from pokemon mystery dungeon 2. pain in the ass. i never beat the guy, and after my 1500th attempt i declared the franchise was awful, the boss was overpowered, and it overall was a horrible sequel to an amazing game.
i don't play pokemon anymore.

Clearly Prototype has made us all much worse gamers by being designed so that the vast majority of the game consists of the player absolutely shitting all over multiple tanks, helicopters and giant beasts so much that whenever an enemy that can actually fight back shows up we consider it hard. For me there was a "that one boss" section but it wasn't so much "that boss" as "that fucking set of missions where you have no powers" but much worse than that, "that one mission where you have to blow up a bloodtox facility to get captured but it's so heavily guarded by supersoldiers and tanks that it takes like an hour to blow it up and you die about 10 times." I was never killed by Elizabeth Greene because there's free health swarming everywhere and basically every one of her attacks could just be avoided by being very far away, then you'd go in and do a bombing run with the giant biomass Kamehameha wave and the body armor activated. I DID, however, find a couple bosses from a long while ago to be much harder:

Both Alma and that ghost samurai armor whose name escapes me from Ninja Gaiden were each a bit of a ***** the first time you had to fight them, before you got used to how shit worked and being abused by a game's difficulty.

Most of the levels on DMD in DMC4 when you're playing Dante's sequence are INSANELY difficult for me because I got way too used to Nero's simpler rhythm. Then Dante's ridiculously complex weapon and combo cycling shows up and throws me for a loop and on the highest difficulty setting I simply can't deal with it. DMC3 was pretty goddamn punishing as well.

That wasn't the problem. The problem was that the ***** took a good half hour or more to beat. It was a crappily designed boss fight, a cake walk that took the piss by being so long. If she could actually fight back it might have been a challenge.

OP: Yeah, Greene, fuck her.

Gannon from OOT. I have no idea why but i just couldn't do it and then just put it off with the occasional attempt at it for like 2 years. That boss drove me nuts.

Hah, that's the first boss! I spent a year on the game and got to Dr. Robotnik and his weird hovercraft. I never got past him...

GOOOOD that fight was torturous. I had a pretty good grasp of the combat system, but that fight is insanely prejudiced against you. His attacks cover half of the room, NOTHING takes off less than a third of your health, but do you know what really kills me about that fight? His soul has taken over the body you were using not five minutes ago, and he somehow manages to be about ten times stronger and have five times more health? How? I guess it shouldn't take more than thirty seconds for the soul of a dying man to adjust to an entirely new body? That was the most unbearably cheap enemy in the series.

That was one of my more enraged entries here, but I actually gave up and looked up the ending on Youtube, and... it's not even that good. *Sigh*

The Archdemon in Dragon Age. For some fucking stupid reason I decided it would be a good idea take him on with Sten, Alistair, and Dog. I still haven't beaten it to this day.

Golbez in the DS version of Final Fantasy IV.
First off, there's a boss battle immediately before he shows up. Once he does, he wipes out everyone in your party except for Cecil. Then Rydia shows up, offering the only hope of defeating him (namely, summons), but also being grievously underleveled.
Almost immediately, she is killed, and reviving her puts Cecil at an extreme risk of dying.

But that's not really what makes it so difficult. The truth is:
a.) Summoning requires MP, and unless you bought ethers before going to the Underworld, you end up very quickly finding yourself without any way of casting magic.
b.) Golbez keeps switching his weakness and affinity, requiring you to cast Libra before attacking.
c.) If he changes affinity while you're waiting to cast a spell, your summon will heal his health by about 3000, undoing whatever damage you've done so far. Worst of all, if you've hit him with one summon already, you're screwed because Rydia, at this point, can only cast two before running out of MP.

I stopped playing this game for about a year because of this battle.

The final boss in Sonic Adventures 2, the Bio-Lizard.

Basically, two hits is death (since there aren't many rings), and he has hundreds of simultaneously hitting homing attacks.

Beat him once. And never will again.

The boss was easy, just stun him (With a stun seed), walk across the room then fire ranged shit.

Also, the sequel was much better than the original IMO. ALOT better.
