I'd say that the most imaginative gun would be the Medigun from TF2. Honestly, a gun that heals people? A gun that helps the world? I've heard it all. (Yawn.)
I can't think of the BEST off the top of my head, but here's a contender: Keening from Morrowind. Kills you in about 2 seconds unless your wearing a special glove, but even though it's damage is pretty standard, it's awesome because it (along with Sunder, a hammer) is the only thing thast can kill the heart of a certain god. So it's creative cause it hates you and powerful because you need it to save the world.
That's my favourite Ryno from any R+C game. 100 shots in a fully automatic missile launcher that completely destroys basically any enemy in less than 5 rockets. And you get 20 missiles in every ammo crate, so you're pretty much fully stocked at all times. The giant Protopet is a joke with this gun.SF_KiLLaMaN said:

I can't remember what its called, but in one of the Destroy all humans! game, a gun shot a pink disk and whatever it hit, (tanks, people,) It just threw them around int he air and slammed them into walls for awhile.
