Two nuclear weapons here: The MIRV from Fallout 3 and the Tactical Nuke from Modern Warfare 2.
Both of these are total game enders.

Best weapon would be the Ryno from Ratchet and Clank Going Commando (it kills anything and everything.)
It's spelled "Ryno" for "Rip you a new one"

Technically, the one in Going Commando is the R.Y.2.N.O., for rip you two new ones. /ratchetandclanknerd

I would have too say the Ryno 5 from Ratchet and Clank A Crack in Time. The reason is that it plays the 1812 Overture when you fire it.

Punch glove on a spring (Can do more damage than a machine gun!)
