Well the Collectors were supposed to be former Protheans, but evolved, or rather, devolved to the point of just being mindless workers for the Reapers, left behind to do their laundry while they take a 50,000 year nap somewhere in deep space. I guess their trying to pick up where Soverign left off, activating the citadel mass relay to get the other reapers out of dark space by building another reaper. I'd guess it looks human because they used humans to make it, like the Reapers used Protheans to make, uh... Reapers. So they have to make a new reaper to activate the mass relay to get the other reapers back, but they also feel like they need to kill Shepard because he/she/it foiled their plans last time so they blew up the Normandy to they spent those two years believing Shepard WAS dead and uh.... Why not just invade our galaxy from Dark Space in the two years while Shepard was inactive, I mean in Mass Effect 1, that was their primary goal and they were determined to get it done.
Well, it's all speculation, I guess. Maybe they'll explain some of this stuff in the next game.
