Sev and Scorch from Star Wars : Republic Commando. The real OTP with special mention to the Boss being a lot more murder happy than one would expect.

Medic from Team Fortress 2. One of the classes I have the least playtime on, still remember more of his lines than the ones I've put most of my time into, plus I now have an insatiable urge to say "Danke Kamerad" at every opportunity.

Cortez from Time Splitters. Be it general funny lines to the eventual "Its time to split" moment it's always a good time with sergeant Cortez.

This is going to sounds a bit strange, but Avitus, the Devastator Marine Sergent from Dawn of War 2. There's just something about his deadpan call of "More corpses" after scoring a kill that makes me smile.

Revolver Ocelot.

He's such an unabashed ham that I find him irresistably entertaining and interesting.

Probably not the answer you are looking for but I always smile when I hear or see one of my favorite game characters ever.
Reaver!
Yes, the loveable asshole from Fable 2 and 3. He's just such a fun character. I honestly can't name another videogame character I cared so much about. They were all just a way to progress, or an obstacle to me. Reaver was actually interesting.
All answers are acceptable so long as they are characters that made you smile. I would be happy with, "That one NPC in the purple shirt in that game I can't remember!" If thinking of them makes you smile with nostalgia than good for you.

I'll post another one, which was mentioned before. Kirby. He's just so cute and fluffy. I got to know him through the old Gameboy classic games, and later through Smash Bro's, Smash Bro's Melee and Kirby: Air Ride. I also still think kirby is one of the most awesome superheroes ever. But that's just me. :)

Bioware companions make me smile all the time. I'm loving all the folks I've picked up so far in DA:I. And I look back on previous Bioware games... There are few characters that don't have at least one line that doesn't make me smile.

Meru from Legend of Dragoon. Though that's mostly nostalgia, the writing in that game is kinda shit. And if I hadn't played it as a kid I'd probably hate it if I tried as an adult. Meru's additions are just the greatest though...

It's probably bad for me to say this but the Arkham-verse Riddler always puts a grin on my face for some reason or another. Perhaps it's just because he's so over-the-top in his performance (I think Wally Wingert has something to do with it for sure) and his delivery is just great. And it's satisfying to see him get his comeuppance
I agree (though, admittedly, I'm biased).

Other than that, there are loads of supercute pokemon (Pikachu, Teddiursa and Squirtle come to mind). And Yoshi. Yoshi is fucking awesome.

The only order that I can put these into would have to be the order that I remember them, not on quality. It's so hard to choose!

First, Claptrap from Borderlands 2 and Borderlands: The Presequel. I love that little guy, and he was the whole reason why I bought Pre-Sequel in the first place. Handsome Jack is up there as well, but not as high.

This next one is one that I just recently experienced, but one that I regardless love. Hoji and Lo Wang from the new Shadow Warrior. Both of them are actually pretty good characters with a good amount of personality and development in the game, and I just like the dynamic that they have going on.

Palutena, Pit, and basically everyone from Kid Icarus: Uprising. This game, dude. It has some amazing writing and lovable characters, and I honestly don't know why I don't play it more often. I like it a lot. It's just really really good based on the plot alone.

Tharja from Fire Emblem: Awakening. Her odd obsession with Robin reaches ludicrous levels to the point that she becomes "normal" for him, which just ends up making her creepier, and I just love every exchange she has with the individual characters.

Neeshka from Neverwinter Nights 2. I don't really know if anyone likes this game, but I can't forget Neeshka, that adorable tiefling rogue that quickly made me love this game. Her instant affection for the main character wouldn't normally interest me, but the fact that she was jealous of every single female I met was adorable in itself, were it not for the fact that she was also a freaking awesome thief and just really cool to talk to. And, I had chosen a tiefling species without knowledge of her existence, so that just made the whole thing even cooler!

Frankly, I just all of these characters so much for so many different reasons that I can't say which one's my favorite. They just all are so good and likable that I can't bear to even think about which one might be better.

Mordin from mass Effect (2 & 3), he was always my favorite recruit. People like to shit on ME3 for the whole ending thing, but as far as a connected narrative ME is a shining example, example; Mordin's singing. In his last scene in ME3 if you managed to make him sing in ME2 then he would in the last scene, but if you didn't, then buh.

Nanako from Persona 4 is so delightfully adorable, you can't help but crack a smile. She's tied with Clementine for the best child character in games for me.

Ok here goes my list of pure gaming awesome:

HK-47 - Irreverent Assasin Droid from Knights of the Old Republic 1 & 2.
Shale - Stone Golem from Dragon Age Origins. Hates birds.
Johnny Gat - Your murder happy right hand man in the Saints Row games.
Andrew Ryan - Ruler of Rapture and antagonist in Bioshock
Minsc - Companion from Baldur's Gate. Spouts nonsense and has a pet "minature giant space hamster".

And yes while I may have three Bioware characters on my list, be grateful I restricted myself to one per game...

Abe from Oddworld: Abe's Oddysee, turning to the camera and shrugging 'I dunno' or rolling into a wall, going oof! and growling in irritation.

Siegmeyer of Catarina from Dark Souls, slumped dejected in his awesome bulky armour, muttering 'still closed, still closed...'

Sir Daniel Fortesque from Medievil, mumbling without his jaw and just being a foppish lanky clown of a protagonist.

Crash bandicoot in CB 3 Time Warp, suffering outlandish but lighthearted loony tunes deaths.

Random park visitor in Jurassic park: Operation genesis, opening up their personal info as a trex eats them and having the tail of the speech bubble info sheet travel down the rex's throat, then stay floating and indicating to the belly where Louise Pikolsky, Age 34, Michigan, now resides.

Fidget, got to be Fidget from Dust: An Elysium Tail. Her one liners, her sarky comments, breaking the fourth wall, she's slightly nuts, full of little easter eggs from other games. Always made me smile while I was playing that vastly underrated game.

There are a few little spoilers in the vid, but its a glimpse into the world of Fidget. I WANT ONE!

Mordin Solus - because he's so manic sometimes but with a science edge. Also his theatrical "easter egg" is pure awesome that puts a huge smile on my face.

Varric and Garrus. I just love both of them and for me they are quintessential video game "bros". Funny, charming and reliable. Any time Garrus covers your back or Varric tells another crazy story I can't help but smile.
And all of this...

Looks like my work here is done!

Ezio Auditore the suave womanizing Assassin who stole every ones heart (and put a blade in those he didn't steal). I also liked Edward Kenway a lot with his attitude towards the whole Templar vs Assassin conflict. And of course his sea shanty singing crew members.

Most Bioware companions as well. Especially in Dragon Age 2, whenever they have a conversation with each other it's pure gold (except for that whiny little ***** Anders). But the cake gets taken by Varric the story telling, crossbow shooting, manly chest haired all around charming dwarf. And Sarcastic Hawke's reactions almost always bring a smile to my face.

I am currently playing Far Cry 4 and longinus is my favorite religious person ever.

If by smiling you mean kind, warm and fuzzy smile, then Yuna from Final Fantasy X. She is easily my favourite lady in all of videogames.

If you mean "haha that was funny" smile, then... hmmm... None, really. I mean, I like humorous characters and maybe sometimes will smile at their jokes, but I need something more to smile simply because of character themself.

I think the main protagonist in the new Tales from Borderlands is the funniest character since Guybrush Threepwood.

HK-47. Anyone who had played KOTOR will know why and for those who haven't play it, here is a video of him-
