Bioshock, followed closely by Spec Ops:The Line. Although both games have an amazing atmosphere, set of twists and variety of themes, Bioshock has a far more interesting and original setting with characters to boot. I'd love to re-experience the first time I entered Rapture >.>.
I was going to say Tales of Symphonia, one of my favourite games of all time. I would also like to play Tales of Vesperia and Alan Wake for the first time once more.

As Zhukov has said, it's pretty much asking me what my favorite games are, but in the form of another question.
I am surprised no one mentioned World of Warcraft.
I had the biggest fun ever playing it for the first time. It truly can't be rivaled by any other game. However: burnout. Now I can't even force myself to play five minutes, as much as I miss the game.
Oh God, I went and made myself sad and longing for it again. Crap.
Ocarina of Time... but I think I'd have to forget much of recent gaming improvements to fully enjoy it.
It was just that feeling of walking out onto Hyrule Field for the first time. It was the first time I really went 'Whoa... this is like a world full of stuff to explore.' Haven't really had that reaction since... well except for Eve Online, but that was tempered with realisation that a lot of work would be involved in just being able to explore.
Honestly surprised that not a single soul has said this yet:
Minecraft
Being able to experience Minecraft again as a total noob would be awesome. The feel of the game when you first start, and the childlike curiosity you experience when you begin that first world... No other game has done that for me yet.
But yeah, Minecraft came to mind almost immediately.
Also, all PS1 Final Fantasy games. They were just amazing when I first played them; following an unpredictable story, the eagerness to see everything resolve itself unexpectedly, the music, finding new secrets...god, I'd love to play those with brand new eyes.

Lots of point and click adventure games such as the first Broken Sword, Grim Fandango, Myst IV: Revelations, etc. Those games have the best stories ever.
Probably Morrowind. I've been able to play through Skyrim multiple times trying out different builds. The first time you play Morrowind, when you don't understand all the mechanics and you're discovering them as you go along, that's the best playthrough you'll have. Once you figure out how to exploit the level up process, it's not really the same. The replay value isn't there when all your characters end up the same.

For one, because that whole journey and experience - and the emotional investment I put into playing it for the first time - was amazing. Second, because of the Extended Cut. I'd rather my first feelings of the ending were "Oh. Well, that's very disappointing", rather than "this is a joke, right? ...Guys?".

Similar reasons to Mass Effect. That game, my god, so much emotional investment. Everything that happens, that feeling of freedom of choice. Obviously I'd play it for the first time again to relive those feelings afresh. But also
I'd like to play it again without the overbearing feeling that none of my choices really matter in the long-run. Not only because a lot of the choices are false dilemmas in the first place, but also since they have little - if any - impact in the long run. Not that it stops me from enjoying the journey, it just dampens it quite a bit for me.
[strong]Fallout 3/NV:[/strong]
I put hundreds of hours into exploring and experimenting with these games. The inclusion of mods means I'm still enjoying them to this day, but I'd really love to recreate the initial feelings I first had with the vanilla games, exploring this big open world with its own little stories.
[strong]Pretty much every game with a twist (MGS, etc.):[/strong]
Pretty obvious. That "HOLY CRAP!" feeling of amazement and shock upon being met with a good twist - it can only happen once. Would be great to be able to relive it.
[strong]Games with puzzles (Portal, Professor Layton, Resident Evil, etc.):[/strong]
Pretty obvious once again, but that feeling of solving a puzzle that you've been struggling with, where your brain finally clicks and figures it out. Especially when it suddenly feels incredibly obvious.
[strong]Scary games (Silent Hill, etc.):[/strong]
The original Silent Hill will never fail to put me at incredible unease. But that tense anxiety, the fear combined with not knowing what's coming - oh lord, that was an experience right there. If I could somehow erase my memory of the game, I'd probably just play it for the rest of my life.
