A level of Duke Nukem (Maybe in Babe Land) where you fight through a crowd of Octobrains towards the end of the level, resurface, and scan the keycard for a door. Then you open it...
2 Assault Enforcers, already a big enough threat on there own, 2 MINI BATTLELORDS, and some MORE cannon fodder enemies.
I was just unbelievably pissed off.

Other wise, I feel bad for saying it so much, encounters in Fallout 3 with metro-ghouls or big ass hordes of Deathclaws.

Playing Penumbra: Overture on hard, carefully making my way towards the frozen lake, checking every corner, listening to every footfall and growl carefully to avoid being spotted. I know the door is down the hallway, but I can't see a damn thing. I flick on my flashlight for an instant, spotting the door, and quickly shut it off again. I carefully start to open the door, and hear rapid footsteps. I whip around just in time to see the glowing eyes of Satan's personal guard dog right up in my face. Bricks were shat.

Penumbra: Black Plague on the first playthrough, near the end with Clarence. Just...just Clarence. Fuck you Clarence.

If you've ever played D&D, the words "rust monster" should explain everything.

Playing a dwarf challenge file on Minecraft with the difficulty set to hard, and then realizing the one spot you've deemed defensible enough to convert into a base is next door to not one, but two dungeons. Based on the sounds, one has spiders and the other has skeletons. You just finished walling yourself in from the rest of the monsters waiting outside in the open cavern. An arrow zips by your head as you turn around to realize that the skeleton dungeon is slightly above you and there's a hole in the ceiling you didn't see before. You notice the hole now because four skeletons just dropped down through it.

In Eternal Darkness, from the GC.

Many many times shit got real there, but it might had something to do with the fact that I was 12 when it got released

Vault 106 in Fallout 3.

I got lost in there and could't find the exit.

And it was just so damn creepy.

Playing though Chibi-Robo, not ******** my pants the same way as the rest of you, but I did the moment I realized I loved this game where I am a little robot cleaning a house.

Most of the enemies in Amnesia: The Dark decent, and numerous moments in Silent Hill 2. Some moments in Silent Hill 1 also had me in fear such as the sewer scene and the carnival.

Playing the Suicide Mission on Mass Effect 2 for the first time.

[SPOILERS}

I had just collapsed the Human Reaper into the dank chasm beneath us, and after a lengthy argument with the Illusive Man, watching that heavily pissed-off reaper claw his way back up the platform.

Pants were s***ed.

The part at the end of Mass Effect 2 where you have to fight the Human-Reaper
Huh, I did not find any part of either Mass Effects scary, apart from maybe the side-mission in Mass Effect 1 where you enter a husk filled room. It was hard, which was what made it terrifying :O.

How do you guys fare in other, what people call 'standard' horror games, such as Amnesia and Dead Space?

Also, for me Thief: Deadly Shadow's, The Cradle mission.

Whatever, just wash your hands.
I got a couple from games that arnt scary, one was in half life 2, I was going around ravenholm for like the 4th playthough and I turned a corner and got the crap surprised out of me by one of those damn black head grabs, the other is in serous sam second encounter HD, I was in a large room with a bunch of pillars and I could hear a kleer somewhere in it, I look all over but still cant find it so I decide to just grab the last ammo pack there and suddenly as I come around one of the pillars I hear it run and it jumps out at me and scares the crap out of me so bad I miss my double barrel blast twice

I've never really played horror games as such, so my examples are more of the "made me jump" variety than outright fear.

1 - Half-Life 1, when you are creeping around some air vents near the start, so you only have a wrench as a weapon, and a head crab jumps right at you.

2 - Half-Life 2, when you are on a roof in a graveyard, and those really fast zombies start jumping all over the place. It's been years, so my memory might be deceiving me, but I think they were making some horrible screaming noises too. I was really low on ammo anyway, so I turned the game off and have never played it since! Shame, because it is probably really good.

Closest I've come to that was in the Department Store of Condemned 1. You go down this little hallway with a display of mannequins off to the left. You move down the hall and have one of Ethan(?)'s little flashes then turn around and suddenly the mannequins are right behind you, and as you move down the hall, they move with you, following.

It was one of the few moments in games that actually SCARED me because at first, I didn't realize that they had changed position and moved closer to me, and then the realization hit me and I just yelled out, "Holy DAMN!" Didn't help that the lights in my house were all off at the time either.

I've never defecated in my pants while I was playing a game.

But... the one time that truly made me go "Oh fuck you!" to a game was in the first FEAR. Twice.

That part in RE4 that makes your TV blow up & do "the ring" style static @ 2 am with all the lights in the house off.
