Time travel. It's been done before really badly, Nintendo has a chance with the technology of the Wii, (or the DS, or a new console. who knows?) to create a game that requires you to travel back in time to recover something else besides stars.

And it consistently flips between 3D, 2D, or 2.5D depending on time period. And hey, add some DS+Wii connectivity (as in... the DS is the time travelling unit Mario uses) and you have a great game!

Well, they could do a game where Bowser is like Freddy Kruger in a way. And sense it's dream land, that opens a door to MILLIONS of ideas.

I'd like to see more 2D Mario. It is the classic formula, and with current generation graphics/physics/level design, it could be fantastic.

Play as Bowser across 8 exotic worlds on your was to kidnap the princess to make more Baby Bowsers. First M rated Mario game ever.

If they were to do another regular Mario game, though, I think it'd be interesting to see what they could do if they had the F.L.U.D.D. in a Galaxy setting (except you could take it off whenever you wanted to).

Galaxy was pretty decent... Yes. It had its flaws, but the gravity themed challenges made for some interesting platforming. It seems like the series has stepped too far away from its roots though. What happened to my fireballs? Where did these cutesy little turtle enemies come from, and what did they do to my vicious armies of Koopa troopas?

Granted, I'm kinda hooked on that game ATM. But I figure Nintendo wouldn't be against ripping off other game ideas. Their cash cow has been re-hashing the same basic game for 24 years now and it hasn't failed yet.
