Depends.
If sexuality was a major part of the game, probably not. That's not bigotry, that's the simple fact that though I am fully for gay rights, I personally am both not gay, and find the idea of men being intimate with other men...awkward and unpleasant. I would fully understand a homosexual not wanting to play a game that thoroughly examines a hetero relationship. If a Homosexual finds straight intimacy awkward and unpleasant, makes total sense to me. Just a matter of persuasion.
If the main character happens to be a gay man?...if it's a good game, sure. And, there doing something different from the norm, so the game designer is obviously willing to take some risks, which is a good thing. I think the world of gaming needs more characters who just happen to be gay. Make it no longer an issue.
....but if it's a flamboyantly gay man, maybe not. Of course, the issue there is flamboyancy, not sexuality.
And if it has Lesbians, that can only be a plus. I know, its a tad primitive of me, but I just can't help it. I'm just hardwired that way.
