I also remember the relief whenever I heard the Safe-Room music in the original Resident Evil, especially after being chased down a corridor by rampaging Zombies when I had no ammo.

At the moment I'm going to say the ringing crack when you get a critical hit in V.A.T.S with the lever action rifle, or Lincoln's repeater in Fallout 3. That sound fills me with glee.
