Well, I beat Resident Evil 5 on the highest non-Professional difficulty setting in one day. Does that count?

Me and my brother came pretty close to beating RE5 on Professional in one day. We went from about Chapter 2 straight on through the end in about 6 hours.

Portal
Half Life 2 Episodes 1 and 2
Gears of War 2. It took the whole day, but come on, I was trapped in my house by 4 feet of snow, I had nothing better to do. it was fun though. co-op campaign on insane is always a blast.
Metroid Prime 3. Yet another long sitting, but what else are you going to do on a rainy day while staying in some weathered old farmhouse in upstate new york?
Tak 2: The Staff of Dreams, a game that has never bored me. I have played that game so many times that I can speedrun it in a few hours with my eyes closed.

I guess you cant really count Left 4 Dead....hmmm Portal was so good i couldnt put it down and I did the same for Ico, which is hands down one of the best games ever made.

Castle Wolfenstein for original Xbox on co-op mode, you can't save in co-op so me and my cousin pulled an all nighter beating it.

Similarly, I also beat Halo 3 in one sitting on co-op mode. At the time I didn't own a console so I managed to borrow one for only one day and rented the game, had no choice to beat it in one go.

Modern Warfare 2, which I say is a good thing for that game. If it was any longer than it would just start repeating the mission types over again and that would just drag down the missions that are already there.

Its a shooter for god sakes, why would you want the main story to be ridiculously long anyway.

The most recent being Bioshock 2. Technically speaking it was two sittings but the first was so short I refuse to count it. The thing is, I only finished it in one sitting because I honestly kept thinking the game was about to end in maybe 20 minutes. Then something would happen, and I was presented with something else I thought would cause the game to end. Etc and so fourth. That was a long day...I was snowed in at the time so I wasn't really going to do anything else anyway.

Lets see. Obviously Portal. Short but challenging. I refused to put that down until I finished it.

Halo:ODST...heck, the first playthrough was a co-op with a friend on the second to last hardest difficulty. Then we did it a while later on legendary. Not very long..or enjoyable..That's a different discussion though.

Other than that, I can't quite think of anything that took more than one sitting from this Gen; Excluding some arcade games.

Legend of Zelda: Twilight Princess. It came out during summer break and I had nothing to do, so I played it incessantly. A full 27 hours later and I'd beaten it and gotten every possible collectible. I then promptly fell asleep.
