I doubt human flesh is a good medium for portals. That or a transdimensional rift is torn open, making a gateway between out universe and that of a nearby universe inhabited by horrific aliens. Then it is up to you to go into the other universe and kill the leader of the aliens. Oh wait, they did that already...

Well even if the portal gun worked on your living tissue for some reason it wouldn't be a big deal. You'd just be intact and have a portal on your skin.
First of all, portals work on any flat surface, it has to be a special surface for it not to work. Secondly, you couldn't really put a portal on yourself due to size problems and the fact that you're probably not flat.
However, assuming that you could place a portal on yourself, in which case you're probably some sort of man behemoth, you could stick you limbs through, and even your body. The portal would be the only thing the couldn't go through, and even if you did attempt it the portal would probably either be impossible to force into the other, or it would automatically shut down, severing your limbs if they were already inside the portal.
As far as I'm concerned, no universal implosion, but you would have to move the portal on you to a different location in order to actually get your whole body through the portal. And for those of you thinking that moving a portal to another location while you're still in it would kill you obviously haven't tried it in-game, what it does is spontaneously teleport the part of you body sticking out of the moved portal to the portal's new location. Not quite sure how that works even in-universe, but that's what happens.
EDIT: Either that or the portal doesn't move, it's been a while since I tried it. In this case you'd simply be unable to actually get your whole body through the portal and be forced to withdraw yourself and move the you-mounted portal.

If the portal works, it's not like it opens a hole in the wall where you can get to the other side. Things that go in the wall come out of a portal on your chest and vice versa. Now if you swallowed a portal. entirely different story. Your organs fall out the other side, and you die.
Alot of people are mentioning that portals can only be placed on special walls but the games setup wasn't quite that. Portals can not nescessarily be placed on everything. But there were several surfaces outside the test chambers of varying material you could place portals on. If you want to go off of the game rules itself, you'd have a hard time since there are no organics for you to shoot at in portal. However there is a way to mod the half-life 2 levels into portal, I did this a while back, found it on some web site. In any case you can essentially play through half life 2, as chell with the portal gun. The Portals will open on just about any surface, it's pretty fun shooting rockets through portals. But they will not open on an organic.
That said, if you COULD open one on an organic theres something important to remember, the portal is on the object, not in it. Your guts wouldn't fall out unless the other portal was inside your body. If the other portal was say on another wall, you'd just have a "hole" on your chest people could enter that room through. Or perhaps the processes for whatever superscience runs the portal gun would just burn you badly and not open a portal, I mean GLaDoS mentions that the portals are safe, but the device is not... then again GlaDoS "enchances the truth" alot.

Maybe the rules chanqe when applying portals to an orqanic, so that the portal scales down to fit on the livinq surface? That could work.
