Mother Nature.
She hates us with a Passion you know.

Day After tomorrow was a warning direct from her!

I'd imagine the fact that we are bound to biological form is a huge problem. You know, since that ends up killing us.

Sandbox Tree.
Climb it. I fucking dare you. [http://cdn-www.cracked.com/articleimages/randall/explode/tree.jpg]

Toxic bark, leaves, and the spikes. The seeds are also, pretty much, frag grenades.

'If this tree could walk, it would enslave humanity. '

I would include disease in a subcategory within the singular enemy we know as nature. The Earth need only to sneeze (metaphorically) to cripple us. landslides, earthquakes, tsunamis, floods, volcanos, ice ages, sink-holes, sandstorms, viruses, microbes...I'm not including animals since my list is already ridiculously long but if headed under 'forces of nature' I think I've flown through a loophole. Besides people can kill bears and sharks. You can't kill a hurricane.
I have to disagree with you on one point.

I'd classify Nature and Life are two completely separate things Nature is ALWAYS Life's biggest threat. So temperature, wind, gravity, and all the natural phenomenon that cause earthquakes, volcanoes, and exploding suns, would be Nature, and would be the biggest threat to Life.

That aside I'd say Viruses are Man's biggest threat. The question is, are Viruses Life or Nature?

Aside from Other humans, what would man's greatest enemy be?

I would have to say in my humble opinion man's greatest enemy would be... Bears...they're godless killing machines.
You take that back!

I'd have to say the laws of physics; gravity and the like. Then again if we didn't have them we'd probably worse off.

The greatest enemy of mankind is physics! Every time we figure out how to best physics and further humanity, it finds another law to slow us down! Physics is the only thing stopping us from taking the stars!

This comment is, of course, a joke.

Edit:

Beat me to it while I was watching robots build cubic structures.

hubris

mankind as a species is sorta full of itself. We have a false sence of entitlement, and feel nature should bow to our wants and needs. We use the planet irresponsibly, with no consideration for the big picture

If we somehow survive ourselves long enough to leave the solar system, any external life we encounter will almost certainly be completely foreign to us. With our current attitudes, we wold most likely see it as more of an animal than another intelligent being. Even if we recognized the intelligence of the other species, our perception of them would cast them as immediately inferior, just because they are not us. This perception will pierce through any diplomatic attempts, which will most likely lead to hostility, and possibly war.
