I don't think it can live up to the original. I'm afraid the humor will be forced and if it is funny it will start another annoying internet meme ("The cake is a lie" is fucking annoying and everyone knows it).
Another concern is about length of the game. Portal 1 was about a 2-4 hours long game which like Yahtzee said is a good length, it prevents it form outstaying its welcome. So How will making Portal longer be to its benefit?
I honestly want to know if others share my concerns. I brought my concerns up to a good friend and he struck me upside the head and told me to NEVER blaspheme against Portal again.

Well there is some stuff you are overlooking. I thought Portal 1 was too short, but I got it for free with Orange Box so I was like oh what's this "Portal" meh might as well give it a try. By the time I finished it, I was like holy crap EPIC!
I also then now went through the entire game again just to find all the extra puzzels and challenges (the radios and the cameras from walls). Also did all the challenge puzzels. The reason behind Yahtzee saying it would outstay its welcome is due to the mechanics of the puzzels (I'm not him but from what I understood this was his logic behind it).
Portal 2 is going to have a whole new set of puzzle mechanics as well as the same basics that made Portal 1 so great. On top of that it has a seperate co-op campaign with some other mechanics that aren't in the SP that are just designed for the co-op.
Overall I think everyone will find that the variety in puzzle designs and the shear mass of puzzle mechanics/elements will mean the game will be fun from start to finish.
I especially can't wait for the co-op, it's double as long as Portal 1 and yeah all those new mechanics, the full time will be used up so to speak as to never bore you as it will always feel fresh.
