Even though she's more than slightly significant I have to go with Aerith from FF7. Thats my answer because as far as I know its the only time that a game has made me full on cry.

The only non-significant one is General Leo from FF3/6 (whatever you call it) because even though he fought for the bad guys he had morals and looked to be switching sides to fight for the good team until...
Kefka backstabs the emperor and kills Leo
I don't understand why people gave a shit that she died. I personally thought she was a bit retarded the way she acted and when she died my reaction was "oh great now I have to use Cait Sith, F***"
Well I was like 6 playing through that game the 1st time lol. I would just use Barret and Cid now

Reminds me of the massive fandom freak-out in Naruto when Hinata was presumably killed by Pain.

I was sad when King Cailan died, the first time I played. It was kinda like "what? you give me a super-idealist character with real power who believes in the truth, and then you're going to kill him right away? Well, there's your George RR Martin moment."

I wasn't at all sad when Duncan died, because it didn't show him dying onscreen, so I expected him to survive and pop up at some point later in the game. He's on the boxart after all, he couldn't just be there for like the first 15 minutes, right? I assumed that he'd get corrupted by "the taint" and return later as a BBEG. I was wrong, but I wasn't quite sure until near the end of the game, when he still hadn't made any cameos, except in Alistair's dialogue. But by then it was far too late to be sad over his death.

MGS: Portable Ops.
In one of the most poignant and moving scenes of the game, the first soldier you recruit, Jonathan, takes a shot for you. It was one of the few redeeming parts of the game but really highlighted the fact that Big Boss was an inspiration to all soldiers[/spoilers]

Luis Sera from Resident Evil 4 springs to mind. He's done some things wrong, he's trying to help;

Ooops.
I get the feeling I was meant to be a lot more surprised by that scene than I actually was.
Nevertheless, a great character who I wish had turned up more often. Particularly as he was voiced by a former Batman (Rino Romano).
Farewell, sweet Luis. Next time I play through that damn section with the house and the zombies coming through the windows I'll think of you.

Forget the dog from Fable 2. I gasped at the death of the dog from Bard's Tale.

But far more then that Amethyst Wind made me remember...

In the older game Sylpheed: The Lost Planet, you will never know anyone's name, you will never see anyone's face; but you will never forget them either.
Faced with the unending hordes of OED your panicked flight and eventual counterattack will be accompanied by death after death from your comrades and I felt every loss.

I salute you...

And so many others...

Your sacrifices were not in vain.

My Buddies in FarCry 2, they had little things about them that made me like them. A few were cocky, some were brutal and some were sympathetic. Never made me feel any better when they died.

Dr. Eli Vance. at least, it was sad to see him go by getting his spine snapped by an Advisor's 4 inch long penis. Not, say, fending off 100 Combine soldiers armed only with a pair of pistols. , a look of grim determination on his face as he stands up to death and kicks it in the crotch before the Combine can even get a shot off. Then he's killed when a nuclear reactor self-destructs, taking along the soldiers and maybe an Advisor.

Saints Row 2: Carlos Mendoza it sucks I couldn't save him.

Fable 2: My dog I named him Dax after my RL dog.

KotOR 2: Kreia or Darth Traya which ever you want to call her. But I guess she wouldn't be in this category would she oh well.

KotOR: Jolee Bindo & Mission Vao I always do the darkside ending so they both have to die.

I've not been affected by a characters death that often in my gaming experience. There were a few from time to time over the years, but most notably (and recently), I'm gonna have to go with a particular character in Half-Life.

The tragic death of Eli Vance.

Been reading Warhammer 40ks Gaunts Ghosts series recently. So many great characters sadly killed off... poor bastards.

I got really attached to all the characters in Valkyria Chronicles, which includes the 40 or so other characters you could pick for your squad. So when they died, I was well and truly crushed. All in all I only lost one as the other deaths were shortly followed by a game over, however there was one death that I couldn't avoid. That was the death of Isara. At first I went into denial, thinking that they couldn't kill her off. Then it hit me and I was well and truly miserable. I have yet to find a game where I have made such an emotional attathment to the characters.

So, so very true. I almost cried when that happened. So unexpected and so damn tragic.
