Ashley from Mass Effect.
I liked her a lot because she was a strong opinionated yet likeable character. She had a great sense of humor, real believable personality, a determined sense of duty and a surprisingly sensitive side as well. Sure she had some faults like her distrust of aliens but she never reaches the level of racism and if encouraged the right way she learns to see things in a different light and becomes more trusting of aliens.
Sure she gets a lot of hate but let me tell you, having played the game twenty times and romanced her every single time I can safely say that the 'crazy redneck religious fanatic who can't stop slurring about aliens' simply put, does not exist.
I see her as a strong, independant woman who has very real flaws but also likeable qualities thrown in. I liked her for that because she felt believable, like an actual human being, not merely a one note damsel in distress who was in awe of Shepard's greatness or some oversexualised femme fatale designed to make me get horny. She was just a believable human character.
Honestly, meaning no offence to any fans of any other ladies in Mass Effect, from where I stand none of them even came close to the level of characterisation I saw in Ashley which is why in both my current playthroughs on Mass Effect 2, Shepard merely contently stared at her picture, awaiting the day he could be reuinited with the woman he loves.

I also love the Arbiter because he's just awesome, a deluded protagonist seeking redemption who ends up leading a revolution once he realises the truth.
