It's really hard to say. Looking back at past GOTY awards, they usually have to be something fresh and new or at least a sequel that really ups the ante.
I think if it meets all the hype, Skyrim has the biggest chance as it's big, epic and different enough from Oblivion to avoid being samey. Oblivion won the majority of GOTY awards in 2006 I believe. Similar deal with Arkham City which seems like a bigger and better sequel of a much-loved game.
It's very unlikely that Uncharted 3 won't be amazing, but I think it will be a touch too similar to Uncharted 2 to win GOTY from a lot of publications, though I have very little doubt that it will win it in some.
Some people have said Mass Effect 3 but that's not out until 2012 I'm afraid. Maybe next year.
That's all for the most expected to win. If all of those games end up somehow not meeting expectations, then I'd say Portal 2 is set to win it at the moment. Would be nice if a new IP comes out and surprises us by being better than all the AAA titles, but very doubtful.
EDIT: Seems most people are doing what their personal GOTY will be. I'm in the camp of "No idea until I've played the rest of the games for this year."
