Also at the begining when theyre in the mansion lobby bit and theyre all like hmm wheres barry or chris trying to be concerned but failing to sound like it :p

Also that corny cast thing in the begining cutscene always cracks me up.
Jill sandwich will always remain the funniest though.

I think the Lucas Arts adventure games get an easy nod for their era. Monkey Island, Full Throttle, Sam and Max etc.

For the Snes-PS1 era, Earthbound had it's charm. RE1 unintentional humor factor was quite high despite how much me and my friends enjoyed and were frightened by it while playing.

The N64-PS2 era had Conker's Bad Fur Day which was hit or miss but really hit with some people.

Current era has Portal which was not necessarily as laugh out loud while you were playing it but when you look back on it and discuss it is really amazingly good.

I actually enjoyed the humor in Bully but being a teenager might be a part of it. A lot of the jokes in it are meant for the teen crowd.

QFT! I was rolling on the ground playing that! Sarcasm is the best! I did play Dungeon Runners for a bit. It was funny for a while. Then It just got old.

Destroy All Humans! is very awesomely written
Monkey Island Series
Timesplitters series(Even the characters are just Epic)
Oblivion(not intentionally) is very easy to make funny

The funniest game I've ever played was World Of Goo thanks to "The Sign Painter" and in the Digital Level where you talk to "MOM".

Now, the funniest is MadWorld because of the announcers.
