As always, my answer is the Rail Driver from Red Faction. For the uninitiated the Rail Driver is a gun with a thermal scope that can no only shoot through anything, including walls, but is also an instant-kill weapon.

My favourite overkill weapon? I know its cheating but Godmode Crowbar on HL2 is really fun. Oh and Archimedes Orbital Laser, I haven't shot it yet as I'm waiting for an awesome moment to use it.

I think using an antimaterial rifle to snipe a molerat can be justified as overkill. (this being my current fav in fallout: nv)
