yes glados is the conciousness of a woman transferred into a robot, but the robot has no gender therefore glados is a genderless villain.

here's one i haven't seen yet:

Dead Rising 2: Case West. and you know, on a much larger, meta kinda way, the entire Dead Rising series' main villian is a female, even though you never meet her until Case West.

Dark Queen from Battletoads. I haven't really scrolled through the entire forum to check if I've been ninja'd or not, but it was the obvious choice.

It's been a while so not 100% if there was a twist at the end but, in Tenchu 2, Kagami was the main villan I think.

Battletoads comes to mind, off the top of my head. And, that one's even better for this example, because, in universe, the Battletoads were game programmers who were warped into a video game of their own creation, which had a female main villian. So, we have a video game that takes place IN a video game, both of which having the same female main villian.

Banjo Kazooie has Grunty, who is female and if you end up losing actually becomes super hot! doesn't exactly motivate a guy to win...

I don't know, Meredith doesn't escape responsibility for what happened. The lyrium idol may have pushed her, but she was a fanatical tyrant way before she ever got her hands on it. Besides, being warped by evil magic doesn't stop her from being female...

Jokes. I can't think of any games not mentioned here, so I won't try. However, I'll make an observation about something I've noticed. While there is a far greater preponderance of male villains, there is one upside for female antagonists; they're all generally pretty unique and reasonably well defined, as opposed to the swarm of generic male bad guys.

So while they're fewer, they're also mostly unique, which is rather a good thing.

Jeane is the last boss, not the main villian :/ their is no main villain.
Jeane is directly responsible for everything that happens in the game and Desperate struggle, by murdering Travis' parents. If she hadn't, Travis wouldn't have become an assassin. If he hadn't became an assassin, Bishop wouldn't have been killed in retribution. If Bishop hadn't been killed, Travis wouldn't have had to kill Batt, destroying Santa Destroy's economy. She seems like the villain to me.
Honestly I think you're reading a far bit into it. Plus Travis became an assassin because he wanted to hook up with Sylvia who he met at a bar~
He explains that he mostly became an assassin for payback. The sex was incidental.

For those that say GLaDOS from the Portal games isn't female, you are right...technically. However, it could be argued that since she has the mind of a female within her and is self aware, she is female. But then you'd have to get into the whole 'what is a human/person/female' thing. I think it would just be easier to call her female and just not think too much about it and not get all philosophical. Even though she is a robot/computer, which is genderless.

As for my contribution to the thread, God of War: Chains of Olympus (it's been mentioned before, just not very well). In it you have to fight Persephone (Greek goddess and queen of the Underworld), and stop her from destroying the world/Mount Olympus... At least I think she's the main villain, it's been a while since I've played it.
