Heh, one more decade and this will apply to Episode 3 as well.

Will Gearbox add their own unique flavour to it? Yes.
Commerical success? Probably.
Acutally good? Who knows...

Duke Nukem was a creation of it's time. It's characterization, storyline, and even concept are dated, even offensive in our current society. That is bound to damage it's image and by proxy, it's sales.
On the other hand, the hype itself might be enough to sell it. The very fact it's been so long between games has provided free marketing and exposure. Heck, even though I probably won't buy it, I would be curious to see how it has aged, and whether it lives up to the expectations of the gaming community.

Most people fully realize 3Drealms did the bulk of the work. Even if it's awful, and I doubt it will be, I can't imagine many will blame Gearbox. If anything people will be thankful Gearbox finally got the damn thing out.
Well Gearbox's name will be on the box, and while the industry may know the story if they release a bad, and poor selling game it will hurt their image. Sort of like "I'm not buying another Bethesda game since New Vegas was so bad" or "I'm not getting Deus Ex because Final Fantasy XIII was really bad".

Also low sales are low sales. No matter whose fault it was.

I think it will do OK, and unless it is a really good game it will sell well, but not set the world on fire.
It highly, highly doubt it will have bad sales (as I mentioned earlier in this thread). The game has been hyped for 12 years, people will buy it just for the novelty of owning. Also, in the odd chance it does preform poorly, it's still likely going to be highly profitable.

That all depends on your meaning of 'success'. If they want to create a good game that will engage the audience then they might have trouble. But if, like most developers, they just want to turn a profit, then that is almost certain.

I think the hype actually hit full circle. By this point, there's not really an expectation that it's going to be amazing, but just curiosity that it took this long to come out.

Plus, after so many realistic military shooters (Call Of Duty, Metal Of Honor) and slightly less realistic sci-fi military shooters (Halo, Half-Life), you have Duke Nukem and Bulletstorm promising to go in the opposite direction with over-the-top action and humor. If the games are both quality, maybe there's a chance for a new trend to emerge out of this.

But back to the OP. Assuming the game is good, I think it will be a solid hit. If the reviewers are really in the mood for this sort of game and it gets amazing reviews, it could actually be a pretty damn big hit. This game is going to have a lot of eyes on it simply because it's been an Internet joke for so long, so reviews and word of mouth (good or bad) are going to have a big influence.

Very true. 2007/8 was the peak of normal hype, 2009 was the peak of "LolNevercummingout" speak, and now 2011 has hit a nice "Holy Shit it's real" equilibrium. Duke's actual release makes a bit of a circle as well. Back then most FPSes had silent, characterless, heroes shooting shit. Then Duke came with a one-liner dispensing actual character to flaunt and have you play as.

In very recent years, especially with Halo having been the defining FPSes for the mainstream up until 2010 or so, got back into the same situation. Now Duke is coming with modern technology and 14 years of perfectionism in his bandolier.

Critic-wise, there will be the stupid and the fair. The stupid being those judge DNF based of it's 14 year development, and not off the actual game. The fair judging it off it's merits as an actual game (Which Yahtzee says he will do). Word of mouth is how Duke 3D got around, so maybe history will repeat itself in a Rule of Three fashion.
